Abstract Uveal melanoma (UM) is a highly metastatic cancer that, in contrast to cutaneous melanoma, largely unresponsive checkpoint immunotherapy. Here, we interrogate the tumor microenvironment at single-cell resolution using scRNA-seq of 59,915 and non-neoplastic cells from 8 primary 3 samples. Tumor reveal novel subclonal genomic complexity transcriptional states. Tumor-infiltrating immune comprise previously unrecognized diversity cell types, including CD8 + T predominantly expressing...

In Brief Objective: To define the prognostic significance of surgical center case volume on outcome for soft tissue sarcoma (STS). Methods: STS cases registered in Florida Cancer Data System (FCDS) between 1981 and 2001 were analyzed. Medical facilities ranked by operative volume. Facilities above 67th percentile defined as high-volume centers (HVCs). Results: Of 4205 identified, 68.1% treated at low-volume (LVCs) 31.9% HVCs. A larger proportion high-grade tumors (53.8% vs. 44.3%) lesions...

Differences in cancer survival based on race, ethnicity, and socioeconomic status (SES) are a major issue. To identify points of intervention improve survival, the authors sought to determine impact for patients with cancers head neck (HN).HN diagnosed between 1998 2002 were examined using linked Florida Cancer Data System Agency Health Care Administration data set.A total 20,915 HN identified, predominantly oral cavity larynx. Overall, 72% male, 89.7% white, 8.4% African American (AA),...

To compare the survival outcomes associated with clinical and pathological response in pancreatic ductal adenocarcinoma (PDAC) patients receiving neoadjuvant chemotherapy (NAC) FOLFIRINOX (FLX) or gemcitabine/nab-paclitaxel (GNP) followed by curative-intent pancreatectomy.Newer multiagent NAC regimens have resulted improved responses PDAC; however, effects of these on remain unknown.Clinicopathological data PDAC treated at 7 academic medical centers were analyzed. This study tested the hypothesis that early routine use of tranexamic acid (TXA) reduces mortality in a subset most critically injured trauma intensive care unit patients.Consecutive patients (n = 1,217) who required emergency surgery (OR) and/or transfusions from August 2009 to January 2013 were reviewed. At surgeon discretion, TXA was administered at median 97 minutes (1-g bolus then 1-g over 8 hours) 150 deemed high risk for hemorrhagic death. With propensity scores based on age, sex,...

Timely hemorrhage control is paramount in trauma; however, a critical time interval from emergency department arrival to operation for hypotensive gunshot wound (GSW) victims not established. We hypothesize that delaying surgery more than 10 minutes increases all-cause mortality patients with GSW.Data of adults (n = 309) hypotension and GSW the torso requiring immediate January 2004 September 2013 were retrospectively reviewed. Patients resuscitative thoracotomies, traumatic brain injury,...

Most often these lesions were seen in females [female-to-male ratio (f/m): 5.5/1]. Hepatic adenomas exclusively FNH predominantly (f/m: 26/1). The optimal long-term regimen for immunosuppression kidney transplant recipients is unknown. We conducted a randomized trial involving 150 to compare tacrolimus/sirolimus, tacrolimus/mycophenolate mofetil (MMF), and cyclosporine/sirolimus. All patients received daclizumab induction maintenance corticosteroids. Median follow-up was 8 yr post-transplant. Acute rejection (AR) occurred significantly less often among those treated with tacrolimus/MMF (12%) than tacrolimus/sirolimus (30%) or...
