BRCA1-BARD1 constitutes a heterodimeric RING finger complex associated through its N-terminal regions. Here we demonstrate that the contains significant ubiquitin ligase activity can be disrupted by breast cancer-derived mutation in BRCA1. Whereas individually BRCA1 and BARD1 have very low activities vitro, combined with exhibits dramatically higher activity. Bacterially purified domains comprising residues 1–304 of 25–189 are capable polymerizing ubiquitin. The steady-state level...
Abstract Deep learning algorithms have been successfully used in medical image classification. In the next stage, technology of acquiring explainable knowledge from images is highly desired. Here we show that deep algorithm enables automated acquisition features diagnostic annotation-free histopathology images. We compare prediction accuracy prostate cancer recurrence using our algorithm-generated with diagnosis by expert pathologists established criteria on 13,188 whole-mount pathology...
LSD1, a lysine-specific histone demethylase, is overexpressed in several types of cancers and linked to poor outcomes. In breast cancer, the significance LSD1 overexpression not clear. We have performed an silico analysis assess relationship expression clinical outcome. demonstrate that prognostic factor especially basal-like subtype cancer with aggressive features. This link also observed samples triple negative cancer. Interestingly, we note correlates down-regulation BRCA1 phenomenon...
Abstract In a substantial number of patients, ductal carcinoma in situ (DCIS) the breast will never progress to invasive carcinoma, and these patients are often overtreated under current clinical criteria. Although various candidate markers available, relevant for delineating risk categories have not yet been established. this study, we analyzed characteristics 431 with DCIS performed whole-exome sequencing analysis 21-patient discovery cohort targeted deep 72-patient validation cohort. We...
Several reports have demonstrated the use of whole‐slide imaging (WSI) for primary pathological diagnosis, but no such studies been published from Asia. We retrospectively collected 1070 WSI specimens 900 biopsies and small surgeries conducted in nine hospitals. Nine pathologists, who participated this study, trained College American Pathologists guidelines, reviewed made diagnoses based on digitized, 20× or 40× optically magnified images with a scanner. After washout interval over 2 weeks,...
Exon 2 of MED12, a subunit the transcriptional mediator complex, has been frequently mutated in uterine leiomyomas and breast fibroadenomas; however, it rarely other tumors. Although mutations were also found leiomyosarcomas, frequency was significantly lower than leiomyomas. Here, we examined MED12 mutation phyllodes tumors, another biphasic tumor with epithelial stromal components related to fibroadenomas. Mutations exon analyzed nine fibroadenomas eleven tumors via Sanger sequencing. A...
The agonistic/antagonistic biocharacter of selective estrogen receptor modulators (SERMs) can have therapeutic advantages, particularly in the case premenopausal breast cancers. Although contradictory effects these been studied terms crosstalk between α (ER) and coactivator dynamics growth factor signaling, molecular basis mechanisms is still obscure. We identify a series regulatory controlling cofactor on ER SERM function, whose activities require F-box protein 22 (Fbxo22). Skp1, Cullin1,...
Background Diffusion-weighted imaging (DWI) and the apparent diffusion coefficient (ADC) are widely used for detecting uterine endometrial cancer. The relationships between ADC values pathological features of cancer have not yet been established. Purpose To investigate whether vary according to histologic tumor cellularity grade. Material Methods We retrospectively reviewed 30 pathologically confirmed cancers. All patients underwent conventional non-enhanced magnetic resonance (MRI) DWI...
Abstract Background Patients with ulcerative colitis (UC) may have bowel urgency (BU), which has been recognised as important due to its links anxiety, depression, and reduced social quality of life. This study examines intestinal ultrasound (IUS) findings associated BU, comparing them endoscopic biomarkers. Methods prospective, single-centre included UC patients undergoing colonoscopy from January September 2023. Transabdominal transperineal IUS was performed, assessing wall thickness...
The minimum graft volume still remains unclear in reduced-size liver transplantation (RLT). This study reports the improved survival of canine RLT using a quarter with aid portahepatic vein shunt (PHVS). In beagles, donor was reduced to right lateral and caudate lobes (quarter graft) or without provision PHVS, transplanted orthotopically recipient. PHVS established by an end-to-end anastomosis portal branch hepatic resected left lobe. Liver chemistries including arterial blood ketone body...
BRCA1 and 53BP1 antagonistically regulate homology-directed repair (HDR) non-homologous end-joining (NHEJ) of DNA double-strand breaks (DSB). The histone deacetylase (HDAC) inhibitor trichostatin A directly inhibits the retention at DSB sites by acetylating H4 (H4ac), which interferes with binding to dimethylated Lys20 (H4K20me2). Conversely, we recently found that BRCA1/BARD1 complex is also affected another methylated residue, H3K9me2, can be suppressed lysine methyltransferase (HKMT)...
Deep learning algorithms have achieved great success in cancer image classification. However, it is imperative to understand the differences between deep and human approaches. Using an explainable model, we aimed compare learning-focused regions of magnetic resonance (MR) images with cancerous locations identified by radiologists pathologists. First, 307 prostate MR were classified using a well-established neural network without locational information cancers. Subsequently, assessed whether...
Dysregulation of homologous recombination (HR) DNA repair has been implicated in breast carcinogenesis and chemosensitivity. Here, we investigated the methylation status sixteen HR genes analyzed their association with tumor subtypes responses to neoadjuvant chemotherapy. Core specimens were obtained before chemotherapy from sixty cases primary cancer following four subgroups: luminal (LBC) pathological complete response (p CR ), LBC stable disease, triple‐negative (TNBC) p TNBC poor...
Abstract Background Invasive lobular carcinoma (ILC) is distinct from invasive ductal (IDC) in terms of their hormonal microenvironments that may require different therapeutic strategies. We previously reported selective estrogen receptor modulator (SERM) function requires F-box protein 22 (Fbxo22). Here, we investigated the role Fbxo22 as a potential biomarker contributing to resistance endocrine therapy ILC. Methods A total 302 breast cancer (BC) patients including 150 ILC were recruited...
We investigated the relationship between ratio of CD4+ to CD8+ T cells (CD4/CD8 ratio) and cell activation, indicated by human leukocyte antigen (HLA)-DR expression, in patients with systemic lupus erythematosus (SLE). found that was decreased SLE this significantly related expression HLA-DR (but not CD4+) cells. These findings may assist understanding pathogenesis SLE. In some patients, CD4/CD8 be good indicators therapeutic efficacy.
OBJECTIVE. The purpose of this study was to describe the features intraductal papilloma on MR ductography using a microscopic coil.
