BRCA1-BARD1 constitutes a heterodimeric RING finger complex associated through its N-terminal regions. Here we demonstrate that the contains significant ubiquitin ligase activity can be disrupted by breast cancer-derived mutation in BRCA1. Whereas individually BRCA1 and BARD1 have very low activities vitro, combined with exhibits dramatically higher activity. Bacterially purified domains comprising residues 1–304 of 25–189 are capable polymerizing ubiquitin. The steady-state level...

Abstract Deep learning algorithms have been successfully used in medical image classification. In the next stage, technology of acquiring explainable knowledge from images is highly desired. Here we show that deep algorithm enables automated acquisition features diagnostic annotation-free histopathology images. We compare prediction accuracy prostate cancer recurrence using our algorithm-generated with diagnosis by expert pathologists established criteria on 13,188 whole-mount pathology...

LSD1, a lysine-specific histone demethylase, is overexpressed in several types of cancers and linked to poor outcomes. In breast cancer, the significance LSD1 overexpression not clear. Abstract In a substantial number of patients, ductal carcinoma in situ (DCIS) the breast will never progress to invasive carcinoma, and these patients are often overtreated under current clinical criteria. Although various candidate markers available, relevant for delineating risk categories have not yet been established. this study, we analyzed characteristics 431 with DCIS performed whole-exome sequencing analysis 21-patient discovery cohort targeted deep 72-patient validation cohort. Several reports have demonstrated the use of whole‐slide imaging (WSI) for primary pathological diagnosis, but no such studies been published from Asia. We retrospectively collected 1070 WSI specimens 900 biopsies and small surgeries conducted in nine hospitals. Nine pathologists, who participated this study, trained College American Pathologists guidelines, reviewed made diagnoses based on digitized, 20× or 40× optically magnified images with a scanner. After washout interval over 2 weeks,...

Exon 2 of MED12, a subunit the transcriptional mediator complex, has been frequently mutated in uterine leiomyomas and breast fibroadenomas; however, it rarely other tumors. Although mutations were also found leiomyosarcomas, frequency was significantly lower than leiomyomas. Here, we examined MED12 mutation phyllodes tumors, another biphasic tumor with epithelial stromal components related to fibroadenomas. Mutations exon analyzed nine fibroadenomas eleven tumors via Sanger sequencing. A...

The agonistic/antagonistic biocharacter of selective estrogen receptor modulators (SERMs) can have therapeutic advantages, particularly in the case premenopausal breast cancers. Although contradictory effects these been studied terms crosstalk between α (ER) and coactivator dynamics growth factor signaling, molecular basis mechanisms is still obscure. We identify a series regulatory controlling cofactor on ER SERM function, whose activities require F-box protein 22 (Fbxo22). Skp1, Cullin1,...

Abstract Background Patients with ulcerative colitis (UC) may have bowel urgency (BU), which has been recognised as important due to its links anxiety, depression, and reduced social quality of life. This study examines intestinal ultrasound (IUS) findings associated BU, comparing them endoscopic biomarkers. Methods prospective, single-centre included UC patients undergoing colonoscopy from January September 2023. Transabdominal transperineal IUS was performed, assessing wall thickness...

BRCA1 and 53BP1 antagonistically regulate homology-directed repair (HDR) non-homologous end-joining (NHEJ) of DNA double-strand breaks (DSB). The histone deacetylase (HDAC) inhibitor trichostatin A directly inhibits the retention at DSB sites by acetylating H4 (H4ac), which interferes with binding to dimethylated Lys20 (H4K20me2). Conversely, we recently found that BRCA1/BARD1 complex is also affected another methylated residue, H3K9me2, can be suppressed lysine methyltransferase (HKMT)...
