The integrative multiomics characterization of minute amounts clinical tissue specimens has become increasingly important. Here, we present an approach called Integral-Omics, which enables sequential extraction metabolites, lipids, genomic DNA, total RNA, proteins, and phosphopeptides from a single biopsy-level specimen. We benchmarked this method with various samples, applied the workflow to perform profiling tissues six patients colorectal cancer, found that tumor exhibited suppressed...

Acute liver injury is commonly caused by bacterial endotoxin/lipopolysaccharide (LPS), and drug overdose such as acetaminophen (APAP). The exact role of epigenetic modification in acute remains elusive. Here, we investigated the histone methyltransferase G9a LPS- or APAP overdose-induced injury. Under D-galactosamine sensitization, liver-specific G9a-deficient mice (L-G9a-/-) exhibited 100% mortality after LPS injection, while control L-G9a+/- littermates showed very mild mortality....

Tumor necrosis factor α (TNFα)‐ and interleukin 1β (IL‐1β)‐induced nuclear factor‐κB (NF‐κB) activation play key roles in inflammation, immunity, cancer development. Here, we identified one of the deubiquitinating enzymes (DUBs), ubiquitin‐specific protease 15 (USP15), as a positive regulator both TNFα‐ IL‐1β‐induced NF‐κB activation. Overexpression USP15 potentiated or IL‐1β‐triggered downstream gene transcription, whereas knockdown had opposite effects. Mechanistically, upon TNFα...

Natural dyes are environmentally friendly and have become important alternatives to synthetic in some dyeing products. Therefore, better extraction techniques need be developed for the process of natural dyes. In this work, cotton fabric was dyed using turmeric solution under an ultralow liquor ratio (materials 1:5) ultrasonic-assisted conditions. Dye different dispersants pH values has been investigated three-factor-three–level Box–Behnken design employed explore It found that sodium...

Coarse wool is a kind of goat that difficult to further process in the textile industry due its large diameter, dispersion, better strength, and less bending. Therefore, coarse often discarded as waste or made into low-cost products. In this work, keratin was extracted from by high-efficiency method, then, an Ag-doped keratin/PA6 composite nanofiber membrane with enhanced filtration antibacterial performance prepared using HCOOH solvent reductant. ABSTRACT Oxidized low‐density lipoprotein (ox‐LDL)‐induced endothelial dysregulation and the abnormal interaction between monocytes have been considered key risk factors for atherosclerosis. The study investigated effects of apelin‐13 on ox‐LDL‐induced dysfunction in human aortic cells (HAECs). Cells were stimulated with ox‐LDL (100 mg/L), either alone or combination at concentrations 3 6 µM. Multiple techniques, including real‐time PCR, Western blot analysis, enzyme‐linked immunosorbent...

Abnormalities of methyl-CpG binding protein 2 (Mecp2) cause neurological disorders with metabolic dysfunction; however, its role in adipose tissues remains unclear. Here, we report upregulated Mecp2 white (WAT) obese humans, as well mice and during vitro adipogenesis. Normal chow-fed adipocyte-specific knockout (Mecp2Adi KO mice) showed a lean phenotype, downregulated lipogenic genes thermogenic that were identified using RNA sequencing. Consistently, the deficiency adipocytes protected from...

Abstract Interleukin-1β (IL-1β)-induced signaling is one of the most important pathways in regulating inflammation and immunity. The assembly receptor complex, consisting ligand IL-1β, IL-1 (IL-1R) type 1 (IL1R1), IL-1R accessory protein (IL1RAP), initiates this signaling. However, how IL1R1-associated complex regulated remains elusive. Angiopoietin like 3 (ANGPTL3), a key inhibitor plasma triglyceride clearance, mainly expressed liver exists both intracellular extracellular secreted forms....
