Abstract We investigated the efficacy of a Wilms' tumor gene 1 (WT1) vaccine combined with gemcitabine (GEMWT1) and compared it (GEM) monotherapy for advanced pancreatic ductal adenocarcinoma (PDAC) in randomized phase II study. randomly assigned HLA-A*02:01– or HLA-A*24:02–positive patients PDAC to receive GEMWT1 GEM. assessed WT1-specific immune responses via delayed-type hypersensitivity (DTH) WT1 peptide tetramer assay detect cytotoxic T lymphocytes (WT1-CTL). Of 91 enrolled, 85 were...

To compare the different stem cell sources used in salvage transplantation for graft failure (GF) after cord blood (CBT), we retrospectively analyzed data of 220 patients who developed GF undergoing CBT between January 2001 and December 2007 underwent a second hematopoietic (HSCT) within 3 months. The donor HSCT were (n = 180), peripheral cells (PBSCs; n 24), bone marrow (BM; 16). cumulative incidence neutrophil engraftment on day 30 was 39% with CB, 71% PBSCs, 75% BM. Multivariate analysis...

Abstract Anti-human leukocyte antigen (HLA) antibodies, particularly donor-specific HLA antibodies (DSA), negatively impact engraftment in hematopoietic cell transplantation. Past studies have proposed various interventions to reduce DSA, but these were primarily from single centers and not large-scale registry data. Therefore, we conducted a retrospective analysis of nationwide data examine the effects anti-HLA on engraftment. Evaluable patients classified into an antibody-negative group (...

Although chemoradiotherapy is considered an efficacious treatment option for patients with locally advanced rectal cancer, the associated radiological toxicities and late anal dysfunction are concerning issues. Herein, we examined efficacy safety of folinic acid, 5-fluorouracil, oxaliplatin, irinotecan (FOLFOXIRI) as upfront therapy marginally unresectable local cancers without distant metastasis. This multicenter, prospective, observational study was designed by Yokohama Colorectal Cancer...

Background Familial hemophagocytic lymphohistiocytosis (FHL) is a rare disease of infancy or early childhood. To clarify the incidence and subtypes FHL in Japan, we performed genetic functional analyses cytotoxic T lymphocytes (CTLs) Japanese patients with FHL. Design Methods Among children (HLH) registered at our laboratory, those more than one following findings were eligible for study entry under diagnosis FHL: positive known mutations, family history HLH, impaired CTL-mediated...

Hematopoietic stem cell transplantation (HSCT) is the only curative treatment for juvenile myelomonocytic leukemia (JMML), but few large studies of HSCT JMML exist. Using data from Japan Society Cell Transplantation registry, we analyzed outcomes 129 children with who underwent between 2000 and 2011. The 5-year overall survival (OS) rate cumulative incidence relapse were 64% 34%, respectively. A regimen busulfan/fludarabine/melphalan was most commonly used (59 patients) provided best...

Hematologic stem cell transplantation (HSCT) is the most potent consolidation therapy for high-risk acute lymphoblastic leukemia (ALL), but their outcomes and complications in adolescent young adult (AYA) patients remain unclear. We compared after HSCT ALL among children (age 1 to 9 years; n = 607), adolescents 10 19 783), adults 20 29 years old, 603), based on Japanese nationwide registry data. The 5-year overall survival (OS) rate AYA was worse than that of children, at 64% (95% confidence...

The effects of allogeneic hematopoietic stem cell transplantation (allo-HSCT) on systemic chronic active Epstein-Barr virus infection (sCAEBV) are yet to be analyzed in a large number patients. Using the Japanese registry database, Transplant Registry Unification Management Program, we investigated outcomes 102 sCAEBV patients who underwent allo-HSCT. median age at HSCT was 21 years, and three-year overall survival (3-year OS) rate 72.5%. Of 90 whose viral load after allo-HSCT evaluated, 56...

Summary Patients with acute megakaryoblastic leukaemia of Down syndrome (DS‐AMKL) have an excellent survival rate; however, patients non‐DS‐AMKL experience poor outcomes. Therefore, this study retrospectively analysed 203 children who underwent their first haematopoietic cell transplantation (HCT) from 1986 to 2015 using a nationwide Japanese HCT registry data assess outcomes for non‐DS‐AMKL. The 5‐year overall (OS) and event‐free (EFS) rates were 43% 38% respectively. OS rate was...

Simpson-Golabi-Behmel syndrome is a congenital malformation associated with mutations in GPC3, which located the Xq26 region. Three new loss-of-function and global X-chromosome rearrangement involving GPC3 were identified. A female sibling of patient, who presented cleft palate hepatoblastoma, carries same chromosomal paradoxical pattern inactivation. These findings support variable alterations, possible mechanism patients.

The breakthrough effects of tyrosine kinase inhibitors (TKIs) have lessened indications for allogeneic hematopoietic stem cell transplantation (HSCT) in chronic myeloid leukemia (CML). However, HSCT is still attractive children and adolescents/young adults (AYAs) requiring lifelong TKI therapy. Nevertheless, little has been reported on the outcomes large clinical studies targeting these age groups. This study aimed to identify prognostic factors HSCT, including reduced-intensity conditioning...
