Identification and structure-guided optimization of a series 4-(pyrazol-4-yl)-pyrimidines as selective CDK4/6 inhibitors is reported herein. Several potency selectivity determinants were established based on the X-ray crystallographic analysis representative compounds bound to monomeric CDK6. A scalable, second-generation synthesis of the densely functionalized eastern tricyclic lactone (+)-6, a common intermediate, for construction nodulisporic acids has been achieved. Modifications to first-generation route now permit access (+)-6 in 17 steps with an overall 16.5% yield. Key carbon−carbon bond constructions include Kirk−Petrow (phenylthio)methylation, Sc(OTf)3-catalyzed hydroxymethylation, Stille carbonylation, and Koga three-component, conjugate addition−alkylation sequence.
