Sclerosteosis is a rare high bone mass genetic disorder in humans caused by inactivating mutations SOST, the gene encoding sclerostin. Based on these data, sclerostin has emerged as key negative regulator of mass. We generated SOST knockout (KO) mice to gain more detailed understanding effects deficiency bone.Gene targeting was used inactivate and generate line KO mice. Radiography, densitometry, microCT, histomorphometry, mechanical testing were characterize impact male female Comparisons...

The p53 tumor-suppressor gene encodes a growth-regulatory protein that has been implicated in programmed cell death. To investigate the possible role of neuronal death, we studied expression associated with excitotoxicity adult rat brain. Within hours systemic administration glutamate analogue kainic acid, mRNA levels were increased neurons exhibiting morphological features damage within kainate-vulnerable brain regions. A similar distribution was found for DNA as evidenced by situ...

T cell activation and tolerance are regulated by costimulatory molecules. Although PD-1 serves as a crucial negative regulator of cells, the function its ligands, PDL1 PDL2, is still controversial. In this study, we created PDL2-deficient mouse to characterize in tolerance. Antigen-presenting cells from PDL2-/- mice were found be more potent vitro over wild-type controls, which depended on PD-1. Upon immunization with chicken ovalbumin, exhibited increased CD4(+) CD8(+) vivo when compared WT...

Background Paclitaxel is commonly used as a cancer chemotherapy drug that frequently causes peripheral neuropathic pain. Inflammasome multiprotein complex consisting of Nod-like receptor proteins (NLRPs), apoptosis-associated speck-like protein, and caspase-1, which functions to switch on the inflammatory process release interleukin-1β. Growing evidences have supported interleukin-1β critical in enhancing paclitaxel-induced However, whether activation NLRP3 inflammasome nerve contributes...

Significance Intrinsic bursts and rhythmic burst discharges are elicited by activation of T-type Ca 2+ channels in the thalamic reticular nucleus (TRN). TRN believed to be critical for generation maintenance thalamocortical oscillations, leading spike-and-wave (SWDs) on cortical electroencephalogram, which hallmarks absence seizures. Using knockout mice that completely lack bursts, however, we show increased tonic firing seems sufficient drug-induced SWD generation. These results call into...

We propose and theoretically demonstrate a method to generate the circularly polarized supercontinuum with three-color electric fields. The field is synthesized from an orthogonally two-color (OTC) laser infrared gating field. All driving pulse durations are extended 40 fs. that imposes curved trajectories for ionized electrons extends time interval between each harmonic emitting. Through adjusting intensity ratios among three components of field, nearly circular isolated attosecond can be generated.
