Biomarkers such as DNA, RNA, and protein are powerful tools in clinical diagnostics therapeutic development for many diseases. Identifying RNA expression at the single cell level within morphological context by situ hybridization provides a great deal of information on gene changes over conventional techniques that analyze bulk tissue, yet widespread use this technique setting has been hampered dearth automated ISH assays. Here we present an version technology RNAscope is adaptable to...

Abstract Intra-tumor heterogeneity (ITH) is a major underlying cause of therapy resistance and disease recurrence, read-out tumor growth. Current genetic ITH analysis methods do not preserve spatial context may detect rare subclones. Here, we address these shortfalls by developing validating BaseScope—a novel mutation-specific RNA in situ hybridization assay. We target common point mutations the BRAF , KRAS PIK3CA oncogenes archival colorectal cancer samples to precisely map morphological...

Since polycystic kidney disease (PKD) was first noted over 30 years ago to have neoplastic parallels, there has been a resurgent interest in elucidating neoplasia-relevant pathways PKD. Taking nontargeted metabolomics approach the B6(Cg)-Cys1(cpk/)J (cpk) mouse model of recessive PKD, we now characterized metabolic reprogramming these tissues, leading glutamine-dependent TCA cycle shunt toward total 2-hydroxyglutarate (2-HG) production cpk compared with B6 wild-type tissue. Oct4 is a known master regulator of stem cell renewal and differentiation. Expression during differentiation regulated by promoter methylation the nucleosome remodeling histone deacetylation (NuRD) complex. Here, we show that Cdk2ap1, negative Cdk2 function cycle, promotes murine embryonic to down-regulate expression. We further this repressor Cdk2ap1 dependent on its physical interaction with methyl DNA-binding protein, Mbd3. Our data support potential molecular link between promoters,...

Feline immunodeficiency virus (FIV) is the domestic cat analogue of HIV infection in humans. Both viruses induce oral disease untreated individuals, with clinical signs that include gingivitis and periodontal lesions. Oral manifestations patients are abated by highly effective combination antiretroviral therapy (cART), though certain persist despite therapy. Microorganisms associated cavity opportunistic infections cause similar pathologies cats. To further develop this model, we evaluated...

Molecular markers defining self-renewing pluripotent embryonic stem cells (ESCs) have been identified by relative comparisons between undifferentiated and differentiated cells. Most of analysis has done under a specific differentiation condition that may present significantly different molecular changes over others. Therefore, it is currently unclear if there are true consensus human ESCs (hESCs). To identify set key genes consistently altered during hESCs regardless conditions, we performed...
