T-cell exhaustion due to persistent antigen stimulation is a key feature of chronic viral infections and cancer. Programmed cell death-1 (PD-1) major regulator exhaustion, blocking the PD-1 pathway restores function improves pathogen control tumor eradication. Immunotherapy targeting inhibitory receptor has demonstrated significant antitumor activity. Recently, antibodies have been approved for use in cancer patients. In this review, we summarize role infection therapeutic potential...
PD-1 is an inhibitory receptor that has a major role in T cell dysfunction during chronic infections and cancer. While demethylation of the promoter DNA observed exhausted cells isolated from chronically infected individuals, little known about when this stable programmed course infection. To assess if impacted by prolonged stimulation effector phase infection, we adoptively transferred virus-specific day 8 CD8 mice with lymphocytic choriomeningitis virus (LCMV) clone 13 into recipient had...
Skin fibrosis in systemic sclerosis (SSc) is accompanied by attrition of dermal white adipose tissue (dWAT) and reduced levels circulating adiponectin. Since adiponectin has potent regulatory effects on fibroblasts, we sought to assess signaling SSc skin biopsies, evaluate mice with gain- loss-of-function mutations. Furthermore, investigated the mechanism action agonist peptides targeting receptors vitro vivo. We found that pathway activity was significantly a subset biopsies. Mice lacking...
In chronic infections and cancer, exhausted CD8 T cells exhibit heterogeneous subpopulations. TCF1+PD-1+ progenitor (Tpex) can self-renew give rise to Tim-3+PD-1+ terminally differentiated that retain their effector functions. Tpex are thus essential maintaining a pool of antigen-specific during persistent antigenic stimulation, only they respond PD-1-targeted therapy. Despite potential as crucial therapeutic target for immune interventions, the mechanisms controlling maintenance...
mTOR has important roles in regulation of both innate and adaptive immunity, but whether how modulates humoral immune responses have yet to be fully understood. To address this issue, we examined the effects rapamycin, a specific inhibitor mTOR, on B cell CD4 T during acute infection with lymphocytic choriomeningitis virus. Rapamycin treatment resulted suppression virus-specific by inhibiting proliferation germinal center (GC) cells. In contrast, number memory cells was increased...
In medical diagnosis, relying on only one type of biomarker is insufficient to accurately identify cancer. Blood-based multicancer early detection can help more than cancer from a single blood sample. this study, super-resolution multispectral imaging nanoimmunosensor (srMINI) based three quantum dots (QDs) different color conjugated with streptavidin was developed for the simultaneous screening various biomarkers in at single-molecule level. experiment, srMINI chip used simultaneously...
Silver double nanorings with circular intra-nanogaps between two of different diameters were synthesized without a linker molecule to confine an incident electromagnetic field in single entity. We used on-demand, rational, and systematic multi-stepwise reactions consisting (1) selective etching gold, (2) rim-on deposition platinum, (3) eccentric growth (4) concentric silver. The resulting silver exhibited high degree homogeneity both shape size, strongly coupled hot zones (or "hot halos",...
Squalene-based emulsion (SE) adjuvants like MF59 and AS03 are used in protein subunit vaccines against influenza virus (e.g., Fluad, Pandemrix, Arepanrix) SARS-CoV-2 Covifenz, SKYCovione). We demonstrate the critical role of uric acid (UA), a damage-associated molecular pattern (DAMP), triggering immunogenicity by SE adjuvants. In mice, elevated DAMP levels draining lymph nodes. Strikingly, inhibition UA synthesis reduced vaccine-induced innate immunity, subsequently impairing optimal...
Background: Co-administration of two or more antimicrobials with anti-anaerobic activity is not recommended except in certain circumstances. We therefore conducted an intervention to reduce unnecessary double anaerobic coverage (DAC) prescription. Materials and Methods:The consisted education using institutional intranet prospective audits feedback provided through collaboration between a pharmacist infectious diseases physician Seoul National University Bundang Hospital, tertiary hospital...
Diabetic kidney disease (DKD) is the leading cause of end-stage disease, and current pharmacological treatment for DKD limited to renin-angiotensin system (RAS) inhibitors. Adenosine detectable in significantly elevated response cellular damage. While all 4 known subtypes adenosine receptors, namely, A1AR, A2aAR, A2bAR, A3AR, are expressed kidney, our previous study has demonstrated that a novel, orally active, species-independent, selective A3AR antagonist, LJ-1888, ameliorates unilateral...
We develop a supersensitive "turn-on format" fluorescence sandwich immunoassay for detecting small single molecules. Gold nanoplate-based biotin antibodies and streptavidin-fluorophores were used instead of streptavidin-horseradish peroxidase reacting with tracer in microplate-based competitive enzyme-linked immunosorbent assay (ELISA). Our platform showed low detection limit 5 zeptomolar (5 × 10-21 M), 5.4 1010 times higher sensitivity than the conventional tune-off format ELISA.
Synthetic strategies of web-above-a-ring (WAR) and web-above-a-lens (WAL) nanostructures are reported. The WAR has a controllable gap between the nanoring core nanoweb with nanopores for effective confinement electromagnetic field in nanogap subsequent surface-enhanced Raman scattering (SERS) dyes inside high signal reproducibility, which attributed to generation circular 3D hot zones along rim Pt@Au nanorings wrapping architecture. More specifically, adopted as plasmonic structural rigidity...
Abstract Graft-versus-host disease (GvHD) is a severe complication of hematopoietic stem cell transplantation driven by activated allogeneic T cells. Here, we identify distinct subset factor-1 (TCF1) + CD8 cells in mouse and xenogeneic transplant models acute GvHD. These TCF1 exhibit characteristics compared to - cells, including lower expression inhibitory receptors higher costimulatory molecules. Notably, the displays exclusive proliferative potential could differentiate into effector upon...
In this study, we investigate the characteristics of two main clones carbapenemase-producing Klebsiella pneumoniae isolates from South Korea, ST11 and ST307, including carbapenem-susceptible isolates. Antibiotic susceptibility, serotype or wzi allelic type, presence virulence genes, with respect to serum resistance macrophage internalization were determined for ST307 had a wide range characteristics, virulence, compared those homogeneous The wzi14 K14 type higher than that other serotypes...
ABSTRACT Adenovirus serotype 5 (Ad5) is one of the most widely used viral vectors and known to generate potent T cell responses. While many previous studies have characterized Ad5-induced CD8 responses, there a relative lack detailed that analyzed CD4 cells elicited by Ad5 vaccination. Here, we immunized mice with encoding lymphocytic choriomeningitis virus (LCMV) glycoprotein (GP) examined GP-specific responses compared them those induced an acute LCMV infection. In contrast infection,...
Abstract Background The recently developed Screening Tool of Older Persons' Prescriptions in Frail adults with a limited life expectancy (STOPPFrail) criteria can be helpful for screening medications (PIMs), but it is yet to widely used clinical practice. Herein, we aimed investigate the prevalence PIMs based on STOPPFrail (STOPPFrail-PIM) among frail older admitted geriatric center. Methods This was retrospective cross-sectional study conducted center at an academic tertiary care hospital...
