Abstract Excessive activation of the TLR4 signalling pathway is critical for inflammation-associated disorders, while negative regulators play key roles in restraining from over-activation. Naringenin a citrus flavonoid with remarkable anti-inflammatory activity, but mechanisms underlying its inhibition LPS/TLR4 are less clear. This study investigated molecular targets and therapeutic effects naringenin vitro vivo . In LPS-stimulated murine macrophages, suppressed expression TNF-α, IL-6,...

Abstract High-throughput biological data analysis commonly involves identifying features such as genes, genomic regions, and proteins, whose values differ between two conditions, from numerous measured simultaneously. The most widely used criterion to ensure the reliability is false discovery rate (FDR), which primarily controlled based on p -values. However, obtaining valid -values relies either reasonable assumptions of distribution or large numbers replicates under both conditions....

Aberrant activation of inflammation signaling triggered by tumor necrosis factor α (TNF-α), interleukin-1 (IL-1), and interleukin-17 (IL-17) is associated with immunopathology. Here, we identify neural precursor cells expressed developmentally down-regulated gene 4-like (NEDD4L), a HECT type E3 ligase, as common negative regulator induced TNF-α, IL-1, IL-17. NEDD4L modulates the degradation mitogen-activated protein kinase 2 (MEKK2) via constitutively directly binding to MEKK2 promotes its...

The effectiveness of immune responses depends on the precision stimulus-responsive gene expression programs. Cells specify which genes to express by activating stimulus-specific combinations stimulus-induced transcription factors (TFs). Their activities are decoded a regulatory strategy (GRS) associated with each response gene. Here, we examined whether GRSs target may be inferred from stimulus-response (input-output) datasets, remains an unresolved model-identifiability challenge. We...
