Abstract The proper regulation of transcription is essential for maintaining genome integrity and executing other downstream cellular functions 1,2 . Here we identify a stable association between the genome-stability regulator sensor single-stranded DNA (SOSS) 3 Integrator-PP2A (INTAC) 4–6 Through SSB1-mediated recognition DNA, SOSS–INTAC stimulates promoter-proximal termination attenuates R-loops associated with paused RNA polymerase II to prevent R-loop-induced instability....

Abstract Recent studies demonstrate the functions of long non-coding RNAs (lncRNAs) in mediating gene expression at transcriptional or translational level. Our previous study identified a Sirt1 antisense (AS) lncRNA transcribed from AS strand. However, its role and regulatory mechanism is still unknown myogenesis. Here, functional analyses showed that overexpression promoted myoblast proliferation, but inhibited differentiation. Mechanistically, was found to activate sense gene, . The...

Brown adipose tissue (BAT) dissipates energy through Ucp1-mediated uncoupled respiration and its activation may represent a therapeutic strategy to combat obesity. Here we show that Lkb1 controls BAT expansion UCP1 expression in mice. We generate adipocyte-specific knockout mice that, compared with wild-type littermates, these exhibit elevated subcutaneous white tissue, have increased mass higher expenditure. Consequently, KO improved glucose tolerance insulin sensitivity, are more resistant...

Hepatic gluconeogenesis is essential for maintaining blood glucose levels during fasting and the major contributor to postprandial hyperglycemia in diabetes. Gluconeogenesis a classic cAMP/protein kinase A-dependent process initiated by glucagon, which elevated In this study, we have shown that p38 mitogen-activated protein (p38) was activated liver primary hepatocytes glucagon or forskolin. Fasting plasma were reduced upon blockade of with either chemical inhibitor small interference RNA...

Enhanced external counterpulsation (EECP) is a circulation assist device that may improve endothelial dysfunction by increasing shear stress. Chronic exposure of vascular cells and smooth muscle to relatively high physiological stress has antiproliferative vasoprotective effects. The present study hypothesizes EECP inhibits intimal hyperplasia atherogenesis modifying stress-responsive gene expression.Thirty-five male pigs were randomly assigned 3 groups: high-cholesterol diet (n=11), plus...

PU.1 is an Ets family transcription factor involved in the myelo-lymphoid differentiation. We have previously demonstrated that also expressed adipocyte lineage. However, expression levels of mRNA and protein preadipocytes do not match mature adipocytes. level higher preadipocytes, whereas its adipocytes but preadipocytes. The underlying mechanism remains elusive. Here, we find miR-155 knockdown or overexpression has no effect on MiR-155 regulates adipogenesis through PU.1, via C/EBPβ which...

Zinc ions highly concentrate in hippocampus and play a key role modulating spatial learning memory. At time when dietary fortification supplementation of zinc have increased the consuming level especially youth, toxicity overdose on brain function was underestimated. In present study, weaning ICR mice were given water supplemented with 15 ppm Zn (low dose), 60 (high dose) or normal lab for 3 months, behavior homeostasis tested. Mice fed high dose showed hippocampus-dependent memory...

Antisense long non-coding RNAs (AS lncRNAs) play important roles in refined regulation of animal gene expression. However, their functions and molecular mechanisms for domestic adipogenesis are largely unknown. Here, we found a novel AS lncRNA transcribed from the porcine PU.1 (also known as SPI1) by strand-specific RT-PCR. Results showed that was expressed generally lower than level mRNA subcutaneous adipose, heart, liver, spleen, lympha, skeletal muscle kidney tissues. We further levels...

microRNAs play an important role in the progression of hepatocellular carcinoma (HCC). In this study, we found that miR-582-5p expression was downregulated hepatoma tissues and HCC cell lines. Upregulation reduced colony number, inhibited cellular proliferation, arrested cycle G0/G1 phase. When inhibited, number increased proliferation were promoted. Further studies showed regulated through directly inhibiting CDK1 AKT3, indirectly cyclinD1.

Abstract Intracellular accumulation of the hyperphosphorylated tau is a pathological hallmark in brain Alzheimer disease. Activation extrasynaptic NMDA receptors (E-NMDARs) induces excitatory toxicity that involved Alzheimer’s neurodegeneration. However, intrinsic link between E-NMDARs and tau-induced neuronal damage remains elusive. In present study, we showed cultured primary cortical neurons activation E-NMDA but not synaptic dramatically increased mRNA protein levels, with simultaneous...

Hepatic lipogenesis is the principal route to convert excess carbohydrates into fatty acids and mainly regulated by two opposing hormones, insulin glucagon. Although stimulates hepatic lipogenesis, glucagon inhibits it. However, mechanism which suppresses remains poorly understood. In this study, we have observed that p38 mitogen-activated protein kinase plays an inhibitory role in lipogenesis. Levels of plasma triglyceride accumulation liver were both elevated when activation was blocked....
