The silicon pixel tracking system for the ATLAS experiment at Large Hadron Collider is described and performance requirements are summarized. Detailed descriptions of detector electronics sensors given. design, fabrication, assembly modules presented. Data obtained from test beams as well studies using cosmic rays also discussed.
During the shutdown of CERN Large Hadron Collider in 2013-2014, an additional pixel layer was installed between existing Pixel detector ATLAS experiment and a new, smaller radius beam pipe. The motivation for this new layer, Insertable B-Layer (IBL), to maintain or improve robustness performance tracking system, given higher instantaneous integrated luminosities realised following shutdown. Because extreme radiation collision rate environment, several radiation-tolerant sensor electronic...
Abstract Specialized or secondary metabolites are small molecules of biological origin, often showing potent activities with applications in agriculture, engineering and medicine. Usually, the biosynthesis these natural products is governed by sets co-regulated physically clustered genes known as biosynthetic gene clusters (BGCs). To share information about BGCs a standardized machine-readable way, Minimum Information Biosynthetic Gene cluster (MIBiG) data standard repository was initiated...
Nonribosomal peptides are chemically and functionally diverse natural products with important applications in medicine agriculture. Bacterial fungal genomes contain thousands of nonribosomal peptide biosynthetic gene clusters (BGCs) unknown function, providing a promising resource for discovery. Core structural features such can be inferred by predicting the substrate(s) adenylation (A) domains synthetases (NRPSs). However, existing approaches to A domain prediction rely on limited datasets...
Fungi are often inconspicuous in nature and this means it is all too easy to overlook their importance.Often referred as the "Forgotten Kingdom", fungi key components of life on planet. The phylum Basidiomycota, considered contain most complex evolutionarily advanced members Kingdom, includes some iconic fungal species such gilled mushrooms, puffballs bracket fungi.Basidiomycetes inhabit a wide range ecological niches, carrying out vital ecosystem roles, particularly carbon cycling symbiotic...
Abstract Semi-synthetic derivatives of the tricyclic diterpene antibiotic pleuromutilin from basidiomycete Clitopilus passeckerianus are important in combatting bacterial infections human and veterinary medicine. These compounds belong to only new class antibiotics for applications, with novel mode action lack cross-resistance, representing a great potential. Basidiomycete fungi, being dikaryotic, not generally amenable strain improvement. We report identification seven-gene gene cluster...
Abstract The rise in antibiotic resistance is a major threat for human health. Basidiomycete fungi represent an untapped source of underexploited antimicrobials, with pleuromutilin—a diterpene produced by Clitopilus passeckerianus —being the only from these leading to commercial derivatives. Here we report genetic characterisation steps involved pleuromutilin biosynthesis, through rational heterologous expression Aspergillus oryzae coupled isolation and detailed structural elucidation...
Trichoderma koningiopsis is a filamentous fungus that produces numerous bioactive molecules and has the potential to be used as biological control plant growth-promoting agent. Here, we present protocol for generating protoplasts of T. koningiopsis. We then describe four techniques stable transformation plasmid DNA: polyethylene glycol (PEG)-mediated protoplasts, Agrobacterium-mediated transformation, electroporation spores, lipofection mycelia. detail procedures confirming expression spCas9...
The surge in antibiotic-resistant Staphylococcus aureus infections has been deemed a major public health concern. There is an urgent need for novel antimicrobial therapies, chemical and nonantibiotic. basidiomycota-derived, secondary metabolite pleurotin shown to be effective against Gram-positive bacteria, while bacteriophages could the ultimate nonantibiotic alternative. In this study, combination of phage K targeting S. was examined. Pleurotin isolated from basidiomycota fungus...
The characterisation of scleric acid, a new natural product from silent and cryptic gene cluster genetically intractable bacteria, its biosynthesis are reported.
, the causal agent of papaya dieback disease, is a devastating pathogen that has caused tremendous decrease in Malaysian export and affected crops neighbouring countries. A few studies on bacterial species capable suppressing
We report two shunt pathways that expand our knowledge on the biosynthesis of antibiotic pleuromutilin. Three new congeners pleuromutilin were isolated, alongside semi-synthesis a novel C-3 methyl, C3–C4 unsaturated analogue.
Pleurotin is a meroterpenoid specialized metabolite made by the fungus Hohenbuehelia grisea, and it lead anticancer molecule due to its irreversible inhibition of thioredoxin-thioredoxin reductase system. Total synthesis pleurotin has been achieved, including through stereoselective route; however, biosynthesis not characterized. In this study, we used isotope-labeled precursor feeding show that nonterpenoid quinone ring congeners derived from phenylalanine. We sequenced genome H. grisea...
Aphids, including the peach-potato aphid, Myzus persicae, are major insect pests of agriculture and horticulture, aphid control measures limited. There is therefore an urgent need to develop alternative more sustainable means control. Recent studies have shown that environmental microbes varying abilities kill insects. We screened a range bacteria isolates for their target species. Tests demonstrated killing aptitude these against six genera (including persicae). No single bacterial strain...
For the development and establishment of concentrating solar thermal collectors a reliable comparable performance testing evaluation is great importance. To ensure consistent in area low- temperature widely accepted commonly used international standard (ISO 9806:2013) already available. In contrast to this, ISO 9806:2013 has not completely penetrated sector yet. On that account detailed literature review been performed on published procedures methodologies as well existing standards. The...
WS9326A is a peptide antibiotic containing highly unusual N-methyl-E-2-3-dehydrotyrosine (NMet-Dht) residue that incorporated during assembly on non-ribosomal synthetase (NRPS). The cytochrome P450 encoded by sas16 (P450Sas) has been shown to be essential for the formation of alkene moiety in NMet-Dht, but timing and mechanism P450Sas-mediated α,β-dehydrogenation Dht remained unclear. Here, we show substrate P450Sas NRPS-associated peptidyl carrier protein (PCP)-bound dipeptide intermediate...
The present work describes the realization of a modular 1-3 kWe, 3-9 kWth micro Combined Heat and Power (m-CHP) system based on innovative Concentrated Solar (CSP) Stirling engine technology. cogeneration energy at distributed level is one leading argument in large part policies related to renewable resources systems. This CSP m-CHP will provide electrical power, heating cooling for single multiple domestic dwellings other small buildings. developed integrates small-scale concentrator optics...
Endophytic fungi constitute a rich source of secondary metabolites that can be manipulated to produce desirable novel analogs for combating current agricultural challenges crop production, especially controlling plant disease. The endophytic fungus Daldinia eschscholtzii MFLUCC 19-0629, was newly isolated from tropical ancient plants, Oncosperma sp., and displays broad-spectrum antifungal antibacterial activities against several pathogens including Ralstonia solanacearum , Fusarium oxysporum...
