Mechanisms underlying severe coronavirus disease 2019 (COVID-19) remain poorly understood. We analyze several thousand plasma proteins longitudinally in 306 COVID-19 patients and 78 symptomatic controls, uncovering immune non-immune linked to COVID-19. Deconvolution of our proteome data using published scRNA-seq datasets reveals contributions from circulating tissue cells. Sixteen percent display reduced inflammation yet comparably poor outcomes. Comparison who died severely ill survivors...

Optogenetic technologies have been the subject of great excitement within scientific community for their ability to demystify complex neurophysiological pathways in central (CNS) and peripheral nervous systems (PNS). The surrounding optogenetics has also extended clinic with a trial ChR2 treatment retinitis pigmentosa currently underway additional trials anticipated near future. In this work, we identify cause loss-of-expression response transdermal illumination an optogenetically active...

Summary COVID-19 has caused over 1 million deaths globally, yet the cellular mechanisms underlying severe disease remain poorly understood. By analyzing several thousand plasma proteins in 306 patients and 78 symptomatic controls serial timepoints using two complementary approaches, we uncover host immune non-immune not previously linked to this disease. Integration of proteomics with nine published scRNAseq datasets shows that SARS-CoV-2 infection upregulates monocyte/macrophage,...

Abstract Background Severe Acute Respiratory Syndrome Coronavirus 2 (SARS-CoV-2) plasma viremia has been associated with severe disease and death in coronavirus 2019 (COVID-19) small-scale cohort studies. The mechanisms behind this association remain elusive. Methods We evaluated the relationship between SARS-CoV-2 viremia, outcome, inflammatory proteomic profiles a of COVID-19 emergency department participants. viral load was measured using qRT-PCR based platform. Proteomic data were...

Abstract Macrophages use an array of innate immune sensors to detect intracellular pathogens and tailor effective antimicrobial responses. In addition, extrinsic activation with the cytokine IFN-γ is often required as well tip scales host-pathogen balance toward pathogen restriction. However, little known about how sensing impacts IFN-γ-activated state. It was observed that in absence IRF3, a key downstream component pathways, IFN-γ-primed macrophages more efficiently restricted bacterium...

Abstract Most patients treated with immune checkpoint blockade (ICB) do not have durable treatment responses. Therefore, there is a critical need to identify early non-invasive biomarkers of response. We performed plasma proteomic analysis (>700 proteins) at three timepoints on 174 metastatic melanoma ICB. leverage independent training and testing cohorts build predictor immunotherapy response that outperforms several tissue-based approaches. found 217 differentially expressed proteins...

In this work, two exercises are described that designed to teach students about the evolution and behavior of electronic bands graphene bilayer graphene. These involve performing extended Hückel molecular orbital theory calculations on polyacenes polycyclic aromatic hydrocarbons. first exercise, investigate how orbitals converge into as polyacene size increases. Further, learn long-range interactions cause frontier-orbital crossing second concepts band structures, crossing, k-space explored...
