Satellite glial cells (SGCs), enveloping primary sensory neurons' somas in the dorsal root ganglion (DRG), contribute to neuropathic pain upon nerve injury. Glial fibrillary acidic protein (GFAP) serves as an SGC activation marker, though its DRG satellite cell specificity is debated. We employed hGFAP-CFP transgenic mouse line, designed for astrocyte studies, explore expression within peripheral nervous system (PNS) after spared injury (SNI). used diverse immunostaining techniques, Western...

Microglia are long-lived cells that constantly monitor their microenvironment. To accomplish this task, they change morphology both in the short and long term under physiological conditions. This makes process of quantifying microglial difficult.By using a semi-manual semi-automatic method to assess fine changes cortical microglia morphology, we were able quantify number, surveillance branch tree starting from fifth postnatal day 2 years life. We identify fluctuating behavior most analyzed...

Abstract Spinal microglia change their phenotype and proliferate after nerve injury, contributing to neuropathic pain. For the first time, we have characterized electrophysiological properties of potential role microglial potassium channels in spared injury (SNI) model We observed a strong increase inward currents restricted at 2 days associated with hyperpolarization resting membrane (RMP) cells compared later time‐points naive animals. identified pharmacologically genetically current as...

The antidiabetic drug metformin has been shown to reduce pain hypersensitivity in preclinical models of chronic and neuropathic humans. Multiple intracellular pathways have described as targets. Among them, is an activator the adenosine 5′-monophosphate protein kinase that can turn modulate activity E3 ubiquitin ligase NEDD4-2 thus post-translational expression voltage-gated sodium channels (NaVs). In this study, we found bulk effect on Na1.7 dependent NEDD4-2. HEK cells,...

(1) Background: As membrane channels contribute to different cell functions, understanding the underlying mechanisms becomes extremely important. A large number of neuronal have been investigated, however, less studied are expressed in glia population, particularly microglia. In present study, we focused on function Kv1.3, Kv1.5 and Kir2.1 potassium both BV2 cells primary microglia cultures, which may impact cellular migration process. Abstract Macrophages and satellite glial cells are found between injured uninjured neurons in the lumbar dorsal root ganglia (DRG). We explored mechanism of neuro‐immune neuron–glia crosstalk leading to hyperexcitability DRG neurons. After spared nerve injury (SNI), CX3CR1 + resident macrophages became activated, proliferated, increased inward‐rectifying potassium channel K ir 2.1 currents. Conditioned medium (CM) by macrophages, obtained from SNI mice, sensitized small naïve mice. However,...

Transient receptor potential melastatin 8 (TRPM8), a membrane ion channel, is activated by thermal and chemical stimuli. In pancreatic ductal adenocarcinoma, TRPM8 required for cell migration, proliferation, senescence associated with tumor size adenocarcinoma stages. Although the underlying mechanisms of these processes have yet to be described, this cation-permeable channel has been proposed as an oncological target. study, glycosylation status was shown affect calcium uptake. expressed in...

The functioning of microglial cells inside the central nervous system depends on their ion channels expression. Microglia are capable synthesizing different cytokines and chemokines, including CXCL1, responding to action via specific receptors. In this study, we explore effect intrathecal injection CXCL1 potassium currents, expressed in CX3CR1-Green Fluorescent Protein labeled microglia transgenic mice. results showed that hyperpolarized by enhancing inward rectifying currents increasing...

Cytokine proteins are involved in different signaling pathways throughout the central nervous system. To study efficacy of an inflammatory cytokine, macrophage migration inhibitory factor (MIF), which acts via several receptor molecules including CXCR2, male rats’ behaviors were determined after intracerebroventricular (ICV) administration MIF. There three treatments: One group received only a second MIF and CXCR2 antagonist (SB265610), third, control carrier medium saline. All rats...
