Point mutations with unclear molecular mechanisms are often associated vancomycin resistance in Staphylococcus aureus. Here, we observed that the walK (G223D) mutation caused decreased expression of genes cell wall metabolism, autolytic activity, thickened walls, and reduced susceptibility.
