Tumour heterogeneity will accumulate and amplify during cell culture passage, even if derived from the same strain. In current study, multiple batches of colorectal cancer HCT116 HT29 lines were obtained using different conditions trypsin digestion processed for RNA sequencing. CD39 was identified as a biomarker highly expressed in easily trypsin-digested cells compared to undigestible ones. Furthermore, determined enhance invasion suppress apoptosis but not affect proliferation. Moreover,...
ABSTRACT Pancreatic cancer is one of the deadliest cancers. Chemokines affect progression pancreatic through various mechanisms. Most CXC chemokine genes, CC genes and CX3C are clustered together within a very short region chromatin. Transcription activity gene clusters usually influenced by chromatin architecture spatial organisation. Nevertheless, chromatin‐mediated regulatory mechanism on transcription has never been studied in cancer. Herein, we determined that expression C‐X‐C motif...
Gastric cancer (GC) is a highly heterogeneous disease, and the response of patients to clinical treatment varies substantially. There no satisfactory strategy for predicting curative effects date. We aimed explore new method efficacy GC based on immune variables detected via flow cytometry. collected 394 tumour tissues from cytometry analysis gating tumour-infiltrating cells (TIICs). Unsupervised consensus clusters were generated cohort classify into different phenogroups, their...

The contribution of RNA processing to tumorigenesis is understudied. Here, we report that the human debranching enzyme (hDBR1), when inappropriately regulated, induces oncogenesis by causing defects, for example, splicing defects. We found wild-type p53 and hypoxia-inducible factor 1 co-regulate hDBR1 expression, insufficient leads a higher rate exon skipping. Transcriptomic sequencing confirmed effect on splicing, metabolite profiling supported observation neoplasm triggered decrease in...
Cancer associated fibroblasts (CAFs) are a crucial cellular component in tumor microenvironment and could promote progression. CAFs usually derived from resident fibroblasts, which undergoing an activated process stimulated by cells. However, the agents mechanism driving this switch have not yet been elucidated. Progranulin (PGRN), well acknowledged secreted glycoprotein, proliferation angiogenesis of colorectal cancer (CRC) cells, high expression PGRN correlated with patient poor prognosis....
Serotonin (5-hydroxytryptamine [5-HT]), a metabolite of tryptophan, acts on the components hypothalamus-hypophysis-gonad axis and induces puberty delay in mammals via 5-HT receptor 1A (HTR1A). However, roles HTR1A hypothalamus pubertal regulation gene expression are not fully understood. In current study, upregulated gonadotropin-releasing hormone (GnRH) GT1-7 GnRH neuronal cells induced by antagonist WAY-100635 maleate was observed vitro. Furthermore, RNA sequencing (RNA-seq) showed...
A variety of neurons in hypothalamus undergo a complicated regulation on transcription activity multiple genes for hypothalamic-pituitary-gonadal axis activation during pubertal development. Identification puberty-associated cell composition and characterization the unique transcriptional signatures across different cells are beneficial to isolation specific advanced understanding their functions. The female Sprague-Dawley rats postnatal day-25, 35 45 were used define dynamic spatial atlas...
Abstract Infantile bullous pemphigoid (BP) is a rare autoantibody‐mediated skin disorder. We report the effective treatment of 6‐month‐old infant with BP using baricitinib, Janus kinase (JAK) inhibitor, after failure steroids and intravenous immunoglobulin. The patient achieved full remission discontinued all medications without any relapses. To our knowledge, this first case baricitinib used in an BP.
An extremely high cancer incidence and the hypersensitivity to DNA crosslinking agents associated with Fanconi Anemia (FA) have marked it be a unique genetic model system study human etiology treatment, which has emerged an intense area of investigation in research. However, there is limited information about relationship between mutated FA pathway development or/and treatment patients without FA. Here we analyzed mutation rates seventeen genes 68 sequence datasets. We found that frequently...

Abstract Growing evidence supports a general hypothesis that aging and cancer are diseases related to energy metabolism. However, the involvement of Fanconi Anemia (FA) signaling, unique genetic model system for studying human or cancer, in metabolism remains elusive. Here, we report FA complementation group D2 protein (FANCD2) functionally impacts mitochondrial ATP production through its interaction with ATP5α, whereas this relationship was not observed mutant FANCD2 (K561R)-carrying cells....
Background: Pulsatile pituitary gonadotropin secretion governed by hypothalamic gonadotropin-releasing hormone (GnRH) is essential for the pubertal onset. The epigenetic mechanism underlying activation of GnRH-dependent regulatory axis in hypothalamus remains elusive. This study aims to explore potential correlation between signature DNA (hydroxyl)methylation and process. Methods: Hypothalamic arcuate nucleus (ARC) mouse at early (4-weeks) late (8-weeks) stages underwent RNA-, RRBS-,...
Abstract Background and Purpose Atopic dermatitis is a common chronic pruritic inflammatory disease of the skin involving neuro‐immune communication. Neuronal mechanism‐based therapeutic treatments remain lacking. We investigated efficacy intravenous lidocaine therapy on atopic underlying mechanism. Experimental Approach Pharmacological intervention, immunofluorescence, RNA‐sequencing, genetic modification immunoassay were performed to dissect basis itch inflammation in dermatitis‐like mouse...
Puberty is the gateway to adult reproductive competence, encompassing a suite of complex, integrative, and coordinated changes in neuroendocrine functions. However, regulatory mechanisms transcriptional reprogramming arcuate nucleus (ARC) during onset puberty are still not fully understood. To understand role epigenetics regulating gene expression, mouse hypothalamic ARCs were isolated at 4 8 weeks, transcriptome, DNA hydroxymethylation, methylation, chromatin accessibility assessed via RNA...
Abstract Background A previous validation study showed a very low sensitivity and higher specificity associated with Hanifin Rajka criteria (H&R) the UK Working Party (UKWP) in diagnosing AD vs. Chinese of atopic dermatitis (AD) for children (CCAD). However, their diagnostic efficacy adult elderly populations remains unknown. Objectives To validate three sets hospital setting. Methods total 1034 patients (aged 19–95 years) from five university dermatological clinics were recruited....
Fanconi anemia (FA) is a rare human genetic disease, resulting from dysfunction in any of 17 known complementation proteins: FANC-A, B, C, D1, D2, E, F, G, I, J, L, M, N, O, P, Q & S, and other unknowns. Besides the severe bone marrow failure, an extremely high incidence cancer as well many clinic symptoms associated with FA patients, cells are insufficiency homologous recombination, DNA mismatch repair, nucleotide excision translesion synthesis, molecular defects, leading to genome...
Objectives Numerous observational studies have reported associations between circulating cytokines and atopic dermatitis (AD); however, the causal relationships them remain unclear. To explore correlations direction of effects AD levels 91 cytokines. Methods Two-sample Mendelian randomization (MR) analyses were conducted to examine using summary statistics from genome-wide association (GWAS). Reverse MR performed investigate reverse causation. Pleiotropy heterogeneity tests assess robustness...
Abstract In recent years, in the development of emerging immunotherapy, B7‐H3 is also termed as CD276 and has become a novel chimeric antigen receptor (CAR)‐T target against glioma other tumours, aroused extensive attention. However, three isoforms (2, 3 4Ig) with controversial expression elusive function tumour especially glioma. The current study mainly focuses on regulatory factors related mechanisms generation different isoforms. First, we have determined that 2Ig dominant high...
