We identified from suckling piglets with diarrhea in China a new bat-HKU2-like porcine coronavirus (porcine enteric alphacoronavirus). The GDS04 strain of this shares high aa identities (>90%) the reported bat-HKU2 strains Coronaviridae-wide conserved domains, suggesting that belongs to same species as HKU2.

Abstract With a possible origin from bats, the alphacoronavirus Porcine epidemic diarrhea virus (PEDV) causes significant hazards and widespread epidemics in swine population. However, ecology, evolution, spread of PEDV are still unclear. Here, 149,869 fecal intestinal tissue samples pigs collected an 11-year survey, we identified as most dominant diarrheal animals. Global whole genomic evolutionary analyses 672 strains revealed fast-evolving genotype 2 (G2) main viruses worldwide, which...

Porcine deltacoronavirus (PDCoV) is a newly identified virus that causes watery diarrhea in newborn piglets and results significant economic losses to the pig industry. Since first reported Hong Kong 2012, PDCoV has been subsequently detected USA, South Korea, Thailand, mainland China. Here we isolated strain of PDCoV, named CHN-GD-2016, from intestinal content diseased piglet with severe farm Guangdong, CHN-GD-2016 could be by immunofluorescence specific rabbit antisera, typical...

Porcine enteric alphacoronavirus (PEAV) was first discovered in China February 2017, and the origin virulence of this novel porcine coronavirus were not fully characterized. Here, we isolated a strain PEAV, named GDS04 that is identified by immunofluorescence typical crown-shaped particles observed with electron microscopy. Genomic analysis reveals PEAV shares close relationship SADS-CoV SeACoV. Furthermore, newborn piglets orally challenged developed clinical symptoms as watery diarrhoea...

Gaussia Luciferase (GLuc) is a renowned reporter protein that can catalyze the oxidation of coelenterazine (CTZ) and emit bright light signal. GLuc comprises two consecutive repeats form enzyme body central putative catalytic cavity. However, deleting C-terminal repeat only limited reduces activity (over 30% residual luminescence intensity detectable), despite being key part How does remaining (tGLuc) CTZ? To address this question, we built structural model tGLuc by removing from resolved...

Since a novel hyper-virulent fowl adenovirus serotype 4 (FAdV-4) infection occurred in 2015, the FAdV-4 has been widely spreading across China, causing significant economic losses to poultry industry. As urgency of issue calls for effective and efficient solutions, present study investigated possibility fiber-2 protein serve as vaccine candidate against FAdV-4. In research, proteins were expressed Escherichia coli, then purified. To evaluate immunogenicity recombinant protein, we both...

Porcine epidemic diarrhea virus (PEDV), Transmissible gastroenteritis (TGEV), deltacoronavirus (PDCoV) and Swine acute syndrome coronavirus (SADS-CoV) rank among the most frequently encountered swine enteric coronaviruses (SECoVs), leading to substantial economic losses industry. The availability of a rapid highly sensitive detection method proves beneficial for monitoring surveillance SECoVs. Based on N genes four distinct SECoVs, novel was developed in this study by combining recombinant...

Porcine epidemic diarrhea virus (PEDV) has caused enormous economic losses to the global pig industry. Currently available PEDV vaccine strains have limited protective effects against variant strains. In this study, highly virulent strain CT was serially passaged in Vero cells for up 120 generations (P120). Characterization of different passages revealed that compared with P10 and P64, P120 had a higher viral titer more obvious cytopathic effects, thereby demonstrating better cell...

African swine fever virus (ASFV) and porcine reproductive respiratory syndrome (PRRSV) infections lead to severe diseases in pigs, resulting significant economic losses for the global industry. While numerous studies have focused on specific gene functions or pathway activities during infection, an investigation of shared immune responses alveolar macrophages (PAMs) after ASFV PRRSV was lacking. In this study, we conducted a comparison using two single-cell transcriptomic datasets generated...

Abstract Swine acute diarrhea syndrome coronavirus (SADS-CoV) is a novel porcine enteric that causes watery diarrhea, vomiting, and dehydration in newborn piglets. The type III interferon (IFN-λ) response serves as the primary defense against viruses replicate intestinal epithelial cells. However, there currently no information available on how SADS-CoV modulates production of IFN-λ. In this study, we utilized IPI-FX cells (a cell line ileum epithelium) an vitro model to investigate...

Porcine reproductive and respiratory syndrome virus (PRRSV) causes severe failure in sows diseases growing finishing pigs results great economic losses to the swine industry. Although vaccines are available, PRRSV remains a major threat pig farms. Thus, there is an urgent need develop antiviral drugs compensate for vaccines.
