In this paper, we attempted to define the role of phagocytosis in virulence Entamoeba histolytica. We have isolated, from a highly phagocytic and virulent strain, clone deficient phagocytosis. Trophozoites wild-type strain HM1:IMSS were fed with Escherichia coli CR34-Thy- grown on 5-bromo,2'-deoxyuridine. The trophozoites that had incorporated base analog through bacteria killed by irradiation 310 nm light. survivors, presumably defective phagocytosis, until log phase submitted two more...

Here, we present evidence that a cysteine protease (EhCP112) and protein with an adherence domain (EhADH112) form the Entamoeba histolytica 112 kDa adhesin. Immunoelectron microscopy immunofluorescence assays using monoclonal antibodies (mAbAdh) revealed that, during phagocytosis, adhesin is translocated from plasma membrane to phagocytic vacuoles. mAbAdh inhibited 54% adherence, 41% 35% 62% destruction of MDCK cell monolayers by live trophozoites their extracts respectively. The cellular bases of the powerful cytolytic activity human protozoan parasite Entamoeba histolytica were explored by studying effect virulent strain HM1:IMSS on epithelial monolayers MDCK cells using a combination time-lapse microcinematography and transmission scanning electron microscopy. Early alterations cell membranes detected measuring changes in transepithelial electrical resistance mounted Ussing chambers. aggressive mechanism E. trophozoites was found to be complex, multifactorial...

Entamoeba histolytica, the protozoan responsible for human amoebiasis, causes between 30,000 and 100,000 deaths per year worldwide. Amoebiasis is characterized by intestinal epithelial damage provoking severe diarrhea. However, molecular mechanisms which this are poorly understood. Here, we studied initial interactions E. histolytica EhCPADH112 virulence complex MDCK Caco-2 cells. By confocal microscopy, discovered that after contact with trophozoites or trophozoite extracts (TE), proteins...

The endosomal sorting complex required for transport (ESCRT) orchestrates cell membrane-remodeling mechanisms in eukaryotes, including endocytosis. However, ESCRT functions phagocytosis(ingestion of ≥ 250 nm particles), has been poorly studied. During intestinal invasion, Entamoeba histolytica opens tight junctions (TJs) reflected by transepithelial electrical resistance (TEER) dropping. To explore the molecular mechanisms underlying this, we studied in vitro and vivo damage produced recombinant E. cysteine protease (rEhCP112) on TJ functions proteins. rEhCP112 reduced TEER Caco-2 cells a dose- time-dependent manner; EhCP112-overexpressing trophozoites provoked major epithelial injury compared to control trophozoites. penetrated...

EhCP112 is an Entamoeba histolytica protease that together with the EhADH112 protein forms EhCPADH complex involved in trophozoite virulence. Here, we produced recombinant and studied its relationships extracellular matrix components target cells. Entamoeba histolytica causes amoebiasis, an infection that kills 100,000 individuals each year. Metronidazole and its derivatives are currently used against this protozoan, but these drugs present adverse effects on human health. Here, we investigated the effect of resveratrol (a natural compound) E. trophozoites viability, as well influence parasite virulence.
