Here, we present evidence that a cysteine protease (EhCP112) and protein with an adherence domain (EhADH112) form the Entamoeba histolytica 112 kDa adhesin. Immunoelectron microscopy immunofluorescence assays using monoclonal antibodies (mAbAdh) revealed that, during phagocytosis, adhesin is translocated from plasma membrane to phagocytic vacuoles. mAbAdh inhibited 54% adherence, 41% 35% 62% destruction of MDCK cell monolayers by live trophozoites their extracts respectively. We cloned 3587...
Entamoeba histolytica, the protozoan responsible for human amoebiasis, causes between 30,000 and 100,000 deaths per year worldwide. Amoebiasis is characterized by intestinal epithelial damage provoking severe diarrhea. However, molecular mechanisms which this are poorly understood. Here, we studied initial interactions E. histolytica EhCPADH112 virulence complex MDCK Caco-2 cells. By confocal microscopy, discovered that after contact with trophozoites or trophozoite extracts (TE), proteins...
The endosomal sorting complex required for transport (ESCRT) orchestrates cell membrane-remodeling mechanisms in eukaryotes, including endocytosis. However, ESCRT functions phagocytosis(ingestion of ≥ 250 nm particles), has been poorly studied. In macrophages and amoebae, phagocytosis is nutrition attack to other microorganisms cells. In Entamoeba histolytica, the voracious protozoan responsible human amoebiasis, phagocytosisis a land mark virulence. Here, we have investigated role ESCRT-III...
During intestinal invasion, Entamoeba histolytica opens tight junctions (TJs) reflected by transepithelial electrical resistance (TEER) dropping. To explore the molecular mechanisms underlying this, we studied in vitro and vivo damage produced recombinant E. cysteine protease (rEhCP112) on TJ functions proteins. rEhCP112 reduced TEER Caco-2 cells a dose- time-dependent manner; EhCP112-overexpressing trophozoites provoked major epithelial injury compared to control trophozoites. penetrated...
EhCP112 is an Entamoeba histolytica protease that together with the EhADH112 protein forms EhCPADH complex involved in trophozoite virulence. Here, we produced recombinant and studied its relationships extracellular matrix components target cells. A DNA fragment containing pro-peptide mature enzyme was expressed bacteria as active (rEhCP112), whereas full gene signal peptide, a non-active protein. The only not expressed. rEhCP112 purified by affinity columns digested azocasein had strong...
Here, we investigated the role of EhVps32 protein (a member endosomal-sorting complex required for transport) in endocytosis Entamoeba histolytica, a professional phagocyte. Confocal microscopy, TEM and cell fractionation revealed cytoplasmic vesicles also located adjacent to plasma membrane. Between 5 30 min phagocytosis, was detected on some erythrocytes-containing phagosomes acidic nature, at 60 it returned appeared images membranous structures vicinity ingested erythrocytes. EhVps32,...
Entamoeba histolytica causes amoebiasis, an infection that kills 100,000 individuals each year. Metronidazole and its derivatives are currently used against this protozoan, but these drugs present adverse effects on human health. Here, we investigated the effect of resveratrol (a natural compound) E. trophozoites viability, as well influence parasite virulence. Trophozoites growth was arrested by 72 μM IC50 determined 220 at 48 h. Cells appeared smaller, rounded in clusters, with...
This paper reports the identification of Ehp53, a p53-like Entamoeba histolytica protein, which binds to human p53 DNA consensus sequence (oli-p53). Monoclonal antibodies against (Ab-1 and Ab-2) recognized single 53 kDa spot in two-dimensional gels inhibited formation complexes produced by E. nuclear extracts oli-p53. Additionally, gene promoter sequences with high homology oli-p53 formed proteins that were abolished Ehp53 protein levels increased UV-irradiated trophozoites. was also...
EhADH112 is an Entamoeba histolytica Bro1 domain-containing protein, structurally related to mammalian ALIX and yeast BRO1, both involved in the Endosomal Sorting Complexes Required for Transport (ESCRT)-mediated multivesicular bodies (MVB) biogenesis. Here, we investigated alternative role MVB protein trafficking pathway by overexpressing 166 amino acids of its N-terminal domain trophozoites. Trophozoites displayed diminished phagocytosis rates accumulated exogenous at cytoplasmic vesicles...
Eukaryotic endocytosis involves multivesicular bodies formation, which is driven by endosomal sorting complexes required for transport (ESCRT). Here, we showed the presence and expression of homologous ESCRT genes in Entamoeba histolytica . We cloned expressed Ehvps4 gene, an member, to obtain recombinant EhVps4 generate specific antibodies, immunodetected cytoplasm trophozoites. Bioinformatics biochemical studies evidenced that rEhVps4 ATPase, whose activity depends on conserved E211...
In higher eukaryotes, mRNA degradation and RNA-based gene silencing occur in cytoplasmic foci referred to as processing bodies (P-bodies). protozoan parasites, the presence of P-bodies their putative role decay have yet be comprehensively addressed. Identification might provide information on how machineries evolved lower eukaryotes. Here, we used immunofluorescence confocal microscopy assays investigate cellular localization proteins human intestinal parasite Entamoeba histolytica found...
The retromer is a cellular structure that recruits and recycles proteins inside the cell. In mammalian yeast, components have been widely studied, but very little in parasites. it formed by SNX-BAR membrane remodeling heterodimer cargo selecting complex (CSC), composed three proteins. One of them, Vps26 protein, possesses flexible intrinsically disordered region (IDR), facilitates interactions with other contributes to binding endosomal membrane. Entamoeba histolytica, protozoan parasite...
Movement and phagocytosis are clue events in colonisation invasion of tissues by Entamoeba histolytica, the protozoan causative human amoebiasis. During phagocytosis, EhRab proteins interact with other functional molecules, conducting them to precise cellular site. The gene encoding EhrabB is located complementary chain DNA fragment containing Ehcp112 Ehadh genes, which encode for EhCPADH complex, involved phagocytosis. This particular genetic organisation suggests that three corresponding...
Entamoeba histolytica is the etiologic agent of human amoebiasis, disease that causes 40,000 to 100,000 deaths annually worldwide. The cytopathic activity as well growth and differentiation this microorganism dependent on both, extracellular free cytoplasmic calcium. However, few known about proteins regulate calcium flux in parasite. In many cells, extrusion from cytosol performed by plasma membrane Ca2+-ATPases calcium/cation exchangers. aim work was identify a exchanger E. analyze its...
Entamoeba histolytica is the protozoan parasite responsible for human amebiasis. It causes up to 100,000 deaths worldwide each year. This has two closely related basal transcription factors, TATA-box binding protein (EhTBP) and TBP-related factor 1 (EhTRF1). TBP binds canonical TATTTAAA-box, as well different TATA variants. TRF1 also TATTTAAA-box. However, their capacity diverse core promoter elements, including GAAC-element, role in gene regulation this remains unknown. EMSA experiments...
