Soma location, dendrite morphology, and synaptic innervation may represent key determinants of functional responses individual neurons, such as sensory-evoked spiking. Here, we reconstruct the 3D circuits formed by thalamocortical afferents from lemniscal pathway excitatory neurons an anatomically defined cortical column in rat vibrissal cortex. We objectively classify 9 cell types estimate number distribution their somata, dendrites, synapses. Somata dendrites most intermingle, while...
Receptor tyrosine kinase (RTK)-mediated activation of downstream effector pathways such as the RAS GTPase/MAP (MAPK) signaling cascade is thought to occur exclusively from lipid membrane compartments in mammalian cells. Here, we uncover a membraneless, protein granule-based subcellular structure that can organize RTK/RAS/MAPK cancer. Chimeric (fusion) oncoproteins involving certain RTKs including ALK and RET undergo de novo higher-order assembly into membraneless cytoplasmic granules...
The broad range of biological responses elicited by transforming growth factor-β (TGF-β) in various types tissues and cells is mainly determined the expression level activity effector proteins Smad2 Smad3. It not fully understood how baseline properties Smad3 are regulated, although this molecule complex with many other at steady state. Here we show that nonactivated Smad3, but Smad2, undergoes proteasome-dependent degradation due to concerted action scaffolding protein Axin its associated...
Expansion microscopy (ExM) increases the effective resolving power of any microscope by expanding sample with swellable hydrogel. Since its invention, ExM has been successfully applied to a wide range cell, tissue, and animal samples. Still, fluorescence signal loss during polymerization digestion limits molecular-scale imaging using ExM. Here, we report development label-retention (LR-ExM) set trifunctional anchors that not only prevent but also enable high-efficiency labeling SNAP CLIP...
Current approaches for surveying whole brains neurons activated during a particular state typically rely on immediate early gene (IEG) expression. However, IEG expression is variable across subjects and brain areas, demanding large sample sizes. Further, it cannot determine if the same or different respond to two events. To overcome these issues, we present whole-brain screening method utilizing transgenic mice label at timepoints. An imaging analysis pipeline surveys activity in ~500 areas...

Abstract Previous studies indicate that the triterpene glycoside actein from herb black cohosh inhibits growth of human breast cancer cells. This study seeks to identify genes altered in cells by treatment with actein, using gene expression analysis. We treated MDA‐MB‐453 at 2 doses, 20 or 40 μg/mL, for 6 24 hr. identified 5 were activated after each treatments are known play a role cellular responses diverse stresses, including DNA damage and unfolded protein responses. In addition, four...
Osteogenesis Imperfecta (OI) is a hereditary connective tissue disorder characterized by bone fragility. IFITM5 gene mutation has been found in all patients with OI type V (< 5% of cases), but its etiologic mechanism remains unclear. OI-V clinical phenotype includes fracture susceptibility variable severity, metaphyseal changes at birth, radial head dislocation, mineralized interosseous membranes, and hyperplasic callus. Short stature, blue sclerae, fractures scoliosis may be rarely observed.
