In this study, the immunoenhancement effect of crude polysaccharides (HLP) from fruiting bodies Helvella leucopus were investigated on immunosuppressive mice model induced by cyclophosphamide (CTX). Monosaccharide composition analysis indicated that HLP was composed mannose, rhamnose and glucose with ratio 8.51:1:1.87. treatment could alleviate atrophy immune organs weight loss observed in mice. Moreover, significantly increased spleen thymus indexes a dose-dependent manner. The...
Intrahepatic cholangiocarcinoma (ICC) is a common primary hepatic tumors with 5-year survival rate of less than 20%. Therefore, it crucial to elucidate the molecular mechanisms ICC. Recently, advance high-throughput chromosome conformation capture (Hi-C) technology help us look insight into three-dimensional (3D) genome structure variation during tumorigenesis. However, its function in ICC pathogenesis remained unclear. Hi-C and RNA-sequencing were applied analyze 3D structures gene...
The Taklamakan Desert is an extreme environment supporting a unique and diverse microbial community with significant potential for exploration. Strain TRM70308T, isolated from desert soil, shares 98.43% 16S rRNA gene sequence similarity Streptomyces alkaliterrae OF1T. Polyphasic taxonomy confirmed TRM70308T as novel species, named xanthus. Genomic analysis revealed that only one of the strain’s 25 biosynthetic clusters (BGCs) formed cluster families (CGFs) within MIBiG database, emphasizing...
Abstract Bacteria have evolved multiple secretion systems for delivering effector proteins into the cytosol of neighboring cells, but roles many these effectors remain unknown. Here, we show that Yersinia pseudotuberculosis secretes an effector, CccR, can act both as a toxin and transcriptional factor. The is secreted by type VI system (T6SS) enter nearby cells same species other (such Escherichia coli ) via cell-cell contact in contact-independent manner. CccR contains N-terminal FIC domain...
Flavonoids have always attracted much attention due to their reversal activity on multidrug resistance (MDR). Eight flavonoids isolated from traditional Chinese medicine Sophora alopecuroides L. were applied test effect MDR associated protein 1 (MRP1) through the established predicting assay. Three flavonostilbenes (alopecurone A, B and D) first found exhibiting potent inhibitory MRP1. All of them dramatically increased 6-carboxyfluorescein diacetate doxorubicin accumulation in...
Four new flavonoids, mortatarins A-D (1-4, resp.), along with eight known flavonoids (5-12) were isolated from the root bark of Morus alba var. tatarica. Their structures established on basis spectroscopic data analysis, and absolute configuration 4 was determined by analysis its CD spectrum. All isolates tested for inhibitory activities against α-glucosidase. Compounds 4, 7, 8 exhibited a significant degree inhibition IC50 values 5.0 ± 0.3, 7.5 0.5, 5.9 0.2 μM, respectively.
The objective of this study was to investigate the biofilm inhibitory activity Streptomyces‐derived actinomycin D against formation by Staphylococcus epidermidis. microtitre plate method and microscopy were used detect S. And an attempt made effect on important components, exopolysaccharides (EPS) in epidermidis using precolumn derivation HPLC. Also cell surface hydrophobicities assessed explore action mechanisms. qPCR performed demonstrate genetic mechanisms Unlike other antibiotics, (1·5...
Chemical investigation of the ethanolic extracts whole plant Ophioglossum pedunculosum afforded seven new homoflavonoid glucosides, pedunculosumosides A−G (1−7). Pedunculosumosides A and C exhibit modest activity blocking HBsAg secretion with IC50 values 238.0 70.5 μM, respectively.
A Streptomyces -like strain, designated TRM 45540T, was isolated from soil of the Loulan area (89° 22′ 22″ E 40° 29′ 55″ N), Xinjiang Province, north-west China, and characterized taxonomically by using a polyphasic study. Phylogenetic analysis 16S rRNA gene sequence revealed that strain 45540T shared 99.87 % similarity with mutabilis NBRC 12800T (GenBank accession number AB184156). The aerobic, Gram-stain-positive, an optimum NaCl concentration for growth 5 (w/v). isolate formed white...
Gentiana veitchiorum Hemsl., Gentianaceae, a traditional Tibetan medicine, was used for the treatment of liver jaundice with damp-heat pathogen, as well headache and chronic pharyngitis. A rapid ultra-performance liquid chromatography, photodiode array detector, quadrupole time-of-flight mass spectrometry method developed fast accurate identification quantification chemical constituents G. veitchiorum. In fact, eighteen compounds were detected identified on basis their spectra, fragment...
Tuberculosis caused by Mycobacterium tuberculosis (M. tb) is a major public health problem with high morbidity and mortality worldwide. In our previous study, we found that fermentation product of Streptomyces flavofungini TRM90047 exhibited anti-M. tb activity decreased the expression level several genes, including rpsL, Rplc ClpC1. Guided heteronuclear single quantum correlation-total correlation spectroscopy (HSQC-TOCSY) fingerprints genome mining, isolated two new 44-membered macrolides,...
Antifungal bioassays led to the isolation of actinomycins D and A1 from Streptomyces luteus TRM45540 collected Norpo in Xinjiang, these compounds were identified by nuclear magnetic resonance spectroscopy. The antifungal activity actinomycin was higher than that A1. Actinomycin clearly inhibited spore germination, hyphal growth biomass accumulation Verticillium dahliae a dose-dependent manner. Flow cytometric analysis with propidium iodide, total ergosterol measurement, cell leakage scanning...
Chemical investigation on 90% ethanol extracts of the aerial parts Glycyrrhiza inflata afforded two new phenolic constituents, 2-(3-methyl-2-butenyl)-3,5,4′-trihydroxy-bibenzyl (1) and (2S)-6-[(E)-3-hydroxymethyl-2-butenyl]-3′,4′,5,7-tetrahydroxy-dihydroflavanone (2) along with seven known dihydroflavanones (3–9). Compounds 1–9 were tested for their minimum inhibitory concentration (MIC) values inhibiting Staphylococcus aureus epidermidis. Compound 1 showed moderate antibacterial activities...
Immunocytes-involved inflammation is considered to modulate the damage in various diseases. Oxidative stress initiated by oxidative agents such as LPS and ROS, which are strongly involved chronic inflammation. Our previous study found that a polysaccharide fraction from Craterellus cornucopioides (CCPP-1) showed good antioxidant activity. However, anti-inflammatory effect of CCPP-1 was still elusive. The objective this evaluate activity its potential mechanism LPS-stimulated RAW264.7...
Three novel actinomycete strains, designated TRM66264-DLMT, TRM88002T and TRM88003T, were isolated by using polyaspartic acid as a water-retaining agent for the enrichment in situ.
