Abstract The robust and stable expression of CD38 in T‐cell acute lymphoblastic leukemia (T‐ALL) blasts makes chimeric antigen receptor (CAR)‐T/natural killer (NK) a potential therapy for T‐ALL. However, normal T/NK cells causes fratricide CAR‐T/NK cells. Here “2‐in‐1” gene editing strategy is developed to generate fratricide‐resistant locus‐specific CD38‐specific CAR integrated into the disrupted locus by CRISPR/Cas9, placed under control either endogenous promoter ( KO/KI ) or exogenous...

Previous research suggests that the care provided to trauma patients could be improved by including early screening and management of emotional distress psychological comorbidity. The Trauma Collaborative Care (TCC) program, which is based on principles well-established models collaborative care, was designed address this gap in center care. Carbapenemase-producing (CP) Enterobacteriaceae are largely responsible for the rapid spread of carbapenem-resistant (CRE). Distinguishing CP-CRE from non-CP-CRE has important infection control implications. In a cohort 198 CRE isolates, isolates that remained susceptible or intermediate to some carbapenem antibiotics, an ertapenem MIC 0.5 μg/ml and meropenem, imipenem, doripenem MICs 2 were best able distinguish isolates.

Abstract T cell genome editing holds great promise to advance a range of immunotherapies but is encumbered by the dependence on difficult‐to‐produce and expensive viral vectors. Here, small double‐stranded plasmid DNA modified mediate high‐efficiency homologous recombination designed. The resulting chimeric antigen receptor (CAR)‐T cells display similar phenotype, transcriptional profile, in vivo potency CAR‐T generated using adeno‐associated vector. As routine testing of clinical isolates for extended-spectrum β-lactamase (ESBL) production (screen plus phenotypic confirmatory testing) is no longer required by the Clinical and Laboratory Standards Institute (CLSI), a number microbiology laboratories use ceftriaxone MICs as proxy means identifying bacteria potential ESBL producers. Data from 1,386 suggest that MIC cutoff 8 μg/ml an excellent predictor production, with positive predictive value negative approaching 100% 99.5%, respectively.
