Summary The lincomycin (LM)‐production gene cluster of the overproducing strain Streptomyces iincolnensis 78‐11 was cloned, analysed by hybridization, as well DNA sequencing, and compared with respective genome segments other producers. lmb/lmr is composed 27 open reading frames putative biosynthetic or regulatory functions ( lmb genes) three resistance (lmr) genes, two which, lmrA lmrC , flank cluster. A very similar overall organization seems to be conserved in four LM producers, although...

The indolizidine alkaloid swainsonine (SW) is a deadly mycotoxin to livestock that can be produced by different plant-associated fungi, including the endophytic entomopathogenic fungi Metarhizium species. SW biosynthetic gene cluster has been identified but genetic mechanism of biosynthesis remains obscure. To unveil pathway, we performed deletions in M. robertsii, heterologous expression core gene, substrate feedings, mass spectrometry, and bioassay analyses this study. It was unveiled via...

Antibiotics biosynthesis is usually regulated by the cluster-situated regulatory gene(s) (CSRG(s)), which directly regulate genes within corresponding biosynthetic gene cluster (BGC). Previously, we have demonstrated that LmbU functions as a regulator (CSR) of lincomycin. And it has been found regulates twenty non-lmb through comparative transcriptomic analysis. However, mode CSRs' targets outside BGC remains unknown.

Lincomycin A is a clinically important antimicrobial agent produced by Streptomyces lincolnensis. In this study, new regulator designated LmbU (GenBank accession no. ABX00623.1) was identified and characterized to regulate lincomycin biosynthesis in S. lincolnensis wild-type strain NRRL 2936. Both inactivation overexpression of lmbU resulted significant influences on production. Transcriptional analysis vivo neomycin resistance (Neor) reporter assays demonstrated that activates expression...

Humans express two ACAT (acyl-CoA:cholesterol acyltransferase) genes, ACAT1 and ACAT2. is ubiquitously expressed, whereas ACAT2 primarily expressed in intestinal mucosa plays an important role cholesterol absorption. To investigate the molecular mechanism(s) responsible for tissue-specific expression of ACAT2, we identified five cis-elements within human promoter, four intestinal-specific transcription factor CDX2 (caudal type homeobox 2), one HNF1α (hepatocyte nuclear 1α). Results...

Lincomycin is one of the most important antibiotics in clinical practice. To further understand regulatory mechanism on lincomycin biosynthesis, we investigated a pleiotropic transcriptional regulator AdpAlin producer S. lincolnensis NRRL 2936. Deletion adpAlin (which generated ΔadpAlin) interrupted biosynthesis and impaired morphological differentiation. We also found that putative AdpA binding sites were unusually scattered promoters all 8 operons biosynthetic gene cluster (BGC). In...

Summary Two different lincomycin‐resistance determinants ( ImrA and ImrB ) from Streptomyces lincolnensis 78–11 were cloned in lividans 66 TK23. The gene was localized on a 2.16 kb fragment, the determined nucleotide sequence of which encoded single open reading frame 1446 bp long. Analysis deduced amino acid suggested presence 12 membrane‐spanning domains showed significant similarities to methylenomycin‐resistance protein (Mmr) coelicolor , QacA Staphylococcus aureus several...

The lincosamide family antibiotic lincomycin is a widely used antibacterial pharmaceutical generated by Streptomyces lincolnensis, and the high-yield strain B48 produces 2.5 g/L lincomycin, approximately 30-fold as wild-type NRRL 2936. Here, genome of S. lincolnensis was completely sequenced, revealing ~10.0 Mb single chromosome with 71.03% G + C content. Lincomycin is one of the most important antibiotics. However, transcriptional regulation network secondary metabolism in Streptomyces lincolnensis, lincomycin producer, remained obscure. AdpA from S. lincolnensis (namely AdpAlin ) has been proved to activate biosynthesis. Here we found that both and melanin took l-tyrosine as precursor, activated biosynthesis well. Three tyrosinases, MelC2, MelD2, MelE, tyrosine peroxygenase, LmbB2, participated different ways. For biosynthesis, MelC2 was...
