Klebsiella pneumoniae is a common cause of sepsis and particularly associated with healthcare-associated infections. New strategies are needed to prevent or treat infections due the emergence multi-drug resistant K. pneumoniae. The goal this study was determine global diversity distribution O (lipopolysaccharide) K (capsular polysaccharide) antigens on large (>500) collection strains isolated from blood inform vaccine development efforts. A total 645 isolates were collected patients...

High-dose isoniazid is recommended to treat multidrug-resistant tuberculosis (MDR TB). Among 958 MDR TB isolates identified in France during 2008-2022, 93.1% exhibited high-level resistance, and molecular testing showed limited diagnostic accuracy predicting resistance. Clinicians should reconsider using high-dose treatment because of suboptimal effect toxicity concerns.

The pathogenesis of diabetic peripheral neuropathy remains uncertain and nonenzymatic glycoxidation is one the contributing mechanisms. aim this study was to assess respective relationship with glycoxidation, compared other identified risk factors, in patients type 2 diabetes. We included 198 diabetes high for vascular complications. Circulating concentrations three advanced glycation end products (carboxymethyllysine, methyl-glyoxal-hydroimidazolone-1, pentosidine) their soluble receptor...

Cancer cell killing might be achieved by the combined use of available drugs. Statins are major anti-hypercholesterolemia drugs, which also trigger apoptosis many cancer types, while docetaxel is a potent microtubule-stabilising agent. Here, we looked at effects lovastatin and in cells. Whole transcriptome microarrays HGT-1 gastric cells demonstrated that strongly suppressed expression genes involved division, had very little transcriptional effects. Both drugs triggered apoptosis, their...

The GeneLEAD VIII (Diagenode, Belgium) is a new, fully automated, sample-to-result precision instrument for the extraction of DNA and PCR detection Mycobacterium tuberculosis complex (MTBC) directly from clinical samples. Deeplex Myc-TB ® assay (Genoscreen, France) diagnostic kit based on deep sequencing 24-plexed amplicon mix allowing simultaneously resistance to 13 antituberculous (antiTB) drugs determination spoligotype. We evaluated performance strategy combining both mentioned tools...

Abstract Background Treatment of multidrug-resistant (MDR) tuberculosis with linezolid is characterized by high rates adverse events. Evidence on therapeutic drug monitoring to predict toxicity scarce. This study aimed evaluate the association trough concentrations severe toxicity. Methods We retrospectively assessed consecutive patients started for MDR between 2011 and 2017. The primary outcome was mitochondrial (SMT) due linezolid, defined as neurotoxicity or myelotoxicity leading...

Context: Calcification of the arterial wall in diabetes contributes to occlusive process occurring below knee. The osteoprotegerin (OPG)/receptor activator nuclear factor κB ligand (RANKL) system is suspected be involved calcification process. Objective: aim study was investigate whether there a link between type 2 and 1) conventional cardiovascular risk factors, 2) serum RANKL OPG levels, 3) neuropathy. Patients Methods: We objectively scored, cross-sectional study, infrapopliteal vascular...

Abstract Background The resistance to all aminoglycosides (AGs) conferred by 16S rRNA methyltransferase enzymes (16S-RMTases) is a major public health concern. Objectives To characterize the genotype, its genetic environment and plasmid support, phylogenetic relatedness of 16S-RMTase-producing Escherichia coli from France. Methods We screened 137 E. isolates resistant clinically relevant AGs nine Parisian hospitals for 16S-RMTases. WGS was performed on clinical with high-level AG (MIC ≥256...

The Clinical and Laboratory Standards Institute recommends the use of Mueller Hinton (MH) medium to perform drug susceptibility testing (DST) Mycobacterium avium complex (MAC) using microdilution method. For MAC, there has been no study on impact media determination minimum inhibitory concentrations (MICs) antibiotics other than clarithromycin. This aimed at determining two used for DST MAC augmenting number pertinent MICs species encountered in clinical practice. treatment infections were...

Ethionamide (ETH) is a second-line antituberculosis drug. ETH resistance (ETH-R) mainly related to the mutations of monooxygenase-activating (EthA), target (InhA), and inhA promoter. Nonetheless, diagnosing ETH-R still challenging. We assessed strategy used for detecting at French National Reference Center Mycobacteria in 497 MDR-TB isolates received from 2008 2016. The genotypic ETH's detection was performed by sequencing ethA, ethR, ethA-ethR intergenic region, promoter multidrug-resistant...
