Antivirals are urgently needed to combat the global SARS-CoV-2/COVID-19 pandemic, supplement existing vaccine efforts, and target emerging SARS-CoV-2 variants of concern. Small molecules that interfere with binding viral spike receptor domain (RBD) host angiotensin-converting enzyme II (ACE2) may be effective inhibitors cell entry. Here, we screened 512 pure compounds derived from natural products using a high-throughput RBD/ACE2 assay identified (-)-hopeaphenol, resveratrol tetramer, in...
Extracellular phospholipases are demonstrated virulence factors for a number of pathogenic microbes. The opportunistic pathogen Candida albicans is known to secrete and these have been correlated with strain virulence. In an attempt clone C. genes encoding secreted phospholipases, Saccharomyces cerevisiae was transformed genomic library screened lipolytic activity on egg-yolk agar plates, traditional screen phospholipase activity. Two identical clones were obtained which exhibited Nucleotide...
Using a mouse model we show that self-complementary (sc) adeno-associated virus (AAV) vectors pseudotyped with capsids of serotypes 2, 7 or 8 induce more potent transgene product-specific CD8+ T cell and antibody responses compared to corresponding single-stranded (ss)AAV vectors. These data suggest the higher rapidly appearing amounts product achieved scAAV may increase detrimental immune in gene transfer recipients.
The erythropoietin-producing hepatocellular carcinoma A3 (EphA3) receptor tyrosine kinase (RTK) regulates morphogenesis during development and is overexpressed mutated in a variety of cancers. EphA3 activation believed to follow 'seeding mechanism' model, which ligand binding the monomeric acts as trigger for signal-productive clustering. We study lateral interactions on surface live cells we demonstrate that forms dimers absence binding. further show these are stabilized by involving...
ABSTRACT Endothelial cells can influence significantly the host inflammatory response against blood-borne microbial pathogens. Previously, we found that endothelial respond to in vitro infection with Candida albicans by secreting interleukin 8 (IL-8) and expressing E-selectin, intercellular adhesion molecule 1 (ICAM-1), vascular cell (VCAM-1). We have now examined mechanisms mediating this response. determined C. stimulated synthesize tumor necrosis factor alpha (TNF-α), which turn induced...
The increasing number of bacteria resistant to combinations β-lactam and β-lactamase inhibitors is creating great difficulties in the treatment serious hospital-acquired infections. Understanding mechanisms structural basis for inactivation these inhibitor-resistant β-lactamases provides a rationale design novel compounds. In present work, SHV-1 Ser130 → Gly variant were inactivated with tazobactam, potent class A inhibitor. Apoenzymes inhibited analyzed by liquid chromatography-electrospray...
Drugs that cure COVID-19 have been marketed; however, this disease continues to ravage the world without becoming extinct, and thus, drug discoveries are still relevant. Since M
ABSTRACT The disruption of a specific gene in Candida albicans is commonly used to determine the function product. We disrupted AAF1 , C. that causes Saccharomyces cerevisiae flocculate and adhere endothelial cells. then characterized multiple heterozygous homozygous mutants. These null mutants adhered cells same extent as did parent organism. However, with presumably genotype revealed significant heterogeneity their growth rates vitro. This was not result transformation procedure per se,...
Quinoxaline derivatization has been the most commonly used approach for sialic acid quantitation of biological samples and glycoproteins by either HPLC or LC-MS/MS.
Portulaca oleracea L. , known as the “vegetable for long life,” is an annual succulent herb that widely distributed worldwide. Many clinical and experimental studies have demonstrated purslane seed (MCXZ) can be used adjunctive alternative therapy treatment of diabetes mellitus (DM). However, underlying active constituents pharmacological mechanisms through which MCXZ exerts effects in DM remain unclear. In present study, we confirmed resulted hypoglycemic activity, lowering fasting blood...
ABSTRACT In this work, a liquid chromatography coupled with electrospray ionization mass spectrometry (LC‐ESI‐MS) method was developed and validated for quantification of bile acids in fecal materials. Co‐eluting matrix impurities materials have been shown to greatly suppress the analytes spectrometry, which is known as effect. To correct large quantitative errors caused by effect, we scheme that combined standard addition internal (SA‐IS). The sample pretreatment involved single step...
Human adenovirus (AdHu)-based candidate AIDS vaccine can provide protection from simian immunodeficiency virus (SIV) transmission and disease progression. However, their potential use may be limited by widespread preexisting immunity to the vector. In contrast, chimpanzee adenoviruses (AdC) is relatively rare. this study, we utilized two regimens of prime-boost immunizations with AdC serotype SAd-V23 (also called AdC6) SAd-V24 AdC7) expressing SIV Gag/Tat test immunogenicity ability protect...
Liquid chromatography/tandem mass spectrometry (LC/MS/MS) is the gold standard of urine drug testing. However, current LC-based methods are time consuming, limiting throughput MS-based testing and increasing cost. This particularly problematic for quantification drugs such as phenobarbital, which often analyzed in a separate run because they must be negatively ionized. This study examined feasibility using dilute-and-shoot flow-injection method without LC separation to quantify with...
CSUOH0901, a novel anticancer derivative of nimesulide, exhibits very promising activities in various cancer cell lines. In order to support further pharmacological and toxicological studies this drug candidate, an LC-MS/MS method was developed validated accordance with the US Food Drug Administration guidelines. The molecules were extracted from plasma samples by protein precipitation then analyzed LC-ESI-MS/MS. An excellent analyte separation achieved using phenomenex C18 column mobile...
Hydrofluoroalkane (HFA) propellants have largely replaced chlorofluorocarbon (CFC) in metered dose inhalers (MDI). It is important to document the pharmacokinetics (PK) and pharmacodynamics (PD) of medications delivered using HFA compared CFC propellants. Six adult asthmatics with mild moderate asthma were selected for study. Each subject inhaled 180 microg albuterol from an MDI holding chamber. Venous blood was collected measuring levels at intervals over 12 h, spirometric measurements...
