Autophagosome-lysosome fusion is a common critical step in various forms of macroautophagy/autophagy including mitophagy, the selective degradation mitochondria. Regulations this process remain poorly defined. Here we have determined role SIGMAR1, unique endoplasmic reticulum membrane protein. Knockout Sigmar1 impaired mitochondrial clearance without altering PINK1-PRKN/Parkin signaling, mouse retinal explants and cultured cells treated with carbonyl cyanide m-chlorophenyl hydrazone (CCCP)...

Osimertinib resistance is an unmet clinical need for the treatment of non-small cell lung cancer (NSCLC), and main mechanism tertiary C797S mutation epidermal growth factor receptor (EGFR). To date, there no inhibitor approved Osimertinib-resistant NSCLC. Herein, we reported a series derivatives as fourth-generation inhibitors which were rationally designed. <title>Abstract</title> Cyclic oligonucleotide-based antiphage signaling systems (CBASS) are widespread bacterial immune that trigger host suicide via cyclic nucleotide-activated effectors. The predominant strategy to induce cell death in CBASS is membrane disruption. Here, we demonstrate patatin-like phospholipase CapV, the most abundant effector, relocates and cleaves phospholipids at pole upon 3’3’-cGAMP binding, inducing polarized disruption death. Using cryo-EM, reveal apo-CapV adopts...

Abstract Background This study aimed to evaluate the association of circular RNA La‐related RNA‐binding protein 4 (circ‐LARP4) with clinical features and prognosis in osteosarcoma patients, further explore its effect on chemosensitivity cells. Methods Seventy‐two patients Enneking stage IIA‐IIB who underwent resection were consecutively enrolled, then, tumor tissues non‐tumor obtained. Circ‐LARP4 tissue/non‐tumor tissue was detected by quantitative polymerase chain reaction. After circ‐LARP4...

Prostate cancer (PCa) is a common male with high incidence and mortality, hormonal therapy as the major treatment for PCa patients troubled by inevitable resistance that makes us identify novel targets PCa. Dual-specificity tyrosine phosphorylation-regulated kinase 2 (DYRK2) was found to be an effective target of PCa, but research on its inhibitors rather little. In this work, potent DYRK2 inhibitor 43 (IC50 = 0.6 nM) acquired through virtual screening structural optimization, which...

T helper (Th) cells are closely involved in vascular inflammation, endothelial dysfunction, and atherogenesis, which the hallmarks of aortic dissection (AD). This study aimed to evaluate clinical value Th1, Th2, Th17 cell measurements Stanford type A AD patients.Stanford patients (N=80) non-AD with chest pain (N = 40) were recruited. Then, peripheral blood CD4+ from all participants detected by flow cytometry. The 30-day mortality was recorded.Th1 higher, while Th2 lower compared (all p <...

The biogenesis of photosystem I (PSI), cytochrome b 6 f (Cytb f) and NADH dehydrogenase (NDH) complexes relies on the spatially temporally coordinated expression translation both nuclear chloroplast genes. Here we report identification regulator 1 (PBR1), a nuclear-encoded RNA-binding protein that regulates concerted NDH, PSI Cytb complexes. We identified Ycf1, one two largest genome-encoded open reading frames as direct downstream target PBR1. Biochemical molecular analyses reveal PBR1 Ycf1...

Aberrant smooth muscle cell (SMC) plasticity is etiological to vascular diseases. Cholesterol induces SMC phenotypic transition featuring high LGALS3 (galectin-3) expression. This proatherogenic process poorly understood for its molecular underpinnings, in particular, the mechanistic role of sterol regulatory-element binding protein-1 (SREBP1), a master regulator lipid metabolism. Herein we show that cholesterol loading stimulated SREBP1 expression mouse, rat, and human SMCs. positively...

The 90-kDa heat shock protein HSP90AA1 is critical for the stability of several proteins that are important tumor progression and thus, a promising target cancer therapy. Selenosemicarbazone metal complexes have been shown to possess anticancer activity through an unknown molecular mechanism. The sigma-2 receptor (S2R) has long been pharmacologically targeted for antipsychotic treatment and tumor imaging. Only recently was it known its coding gene role implicated in cholesterol homeostasis. Here, we have investigated the transcriptional control of S2R by Bromo/ExtraTerminal epigenetic reader family (BETs, including BRD2, 3, 4) upon perturbation. Cholesterol deprivation induced ARPE19 cells using a blocker lysosomal export. This condition up-regulated mRNA protein, also SREBP2 but...

Obesity is increasing at epidemic rates across the US and worldwide, as are its co-morbidities, including type-2 diabetes cardiovascular disease. Thus, targeted interventions to reduce prevalence of obesity utmost importance. The sigma-1 receptor (S1R) sigma-2 (S2R; encoded by Tmem97) belong same class drug-binding sites, yet they genetically distinct. There multiple ongoing clinical trials focused on sigma receptors, targeting diseases ranging from Alzheimer’s disease through chronic pain...
