Bisphosphonates are effective agents for the management of osteoporosis. Their low bioavailability and potency necessitate frequent administration on an empty stomach, which may reduce compliance. Gastrointestinal intolerance limits maximal dosing. Although intermittent intravenous treatments have been used, optimal doses dosing interval not systematically explored.

Growing evidence suggests that vitamin D deficiency may be one of the most important environmental factors for development multiple sclerosis (MS).The objectives this paper are to evaluate serum 25-hydroxyvitamin (25(OH)D) levels in patients with clinically isolated syndromes (CIS) and examine whether they related MS risk. This is a retrospective study 100 CIS hospitalized from 2000 2009 at San Raffaele Hospital, Milan, Italy. We evaluated baseline 25(OH)D level as well clinical, brain...

Transgenic mice over-expressing calcitonin gene-related peptide (CGRP) in osteoblasts have increased bone density due to formation, thus suggesting that CGRP plays a role metabolism. In this study we determined the relationship between CGRP, canonical Wnt signaling and apoptosis human (hOBs) consideration of well-documented involvement pathway cells. Primary cultures hOBs were treated with 10(-8) M. Levels β-catenin, which is cytoplasmic protein mediator signaling, mRNA determined. increases...

The calcitonin peptides [calcitonin (CT), gene-related peptide (CGRP), amylin] share many biological actions, including activity on bone cells. In the present study, CT (10 −11 to 10 −9 M) stimulated [ 3 H]thymidine incorporation in primary cultures of human osteoblasts (hOB), as already demonstrated for CGRP and amylin. RT-PCR analysis showed that receptor receptor-like are both expressed hOB. these cells, −10 amylin M), contrast −8 did not increase cAMP production. All three protein kinase...

To evaluate the potential role of quantitative ultrasound (QUS) investigation in assessing osteopenia prematurity.QUS parameters measured at time discharge were related to anthropometric characteristics and age (postnatal gestational) 51 (34 female 17 male) preterm infants fed fortified human milk.QUS evaluation was performed humerus (h) by measuring two parameters: velocity (hSOS, m/s) bone transmission (hBTT, micros). The uPAR and its ligand uPA are expressed by both osteoblasts osteoclasts. Their function in bone remodeling is unknown. We report that uPAR-lacking mice display increased BMD, osteogenic potential of osteoblasts, decreased osteoclasts formation, altered cytoskeletal reorganization mature osteoclasts.Urokinase receptor (uPAR) actively involved the regulation important cell functions, such as proliferation, adhesion, migration. It was previously shown major players remodeling, osteoclasts,...

Bone fragility and associated fracture risk are major problems in aging. Oxidative stress mitochondrial dysfunction play a key role the development of bone fragility. Mitochondrial is closely with excessive production reactive oxygen species (ROS). L-Carnitine (L-C), fundamental cofactor lipid metabolism, has an important antioxidant property. Several studies have shown how L-C enhances osteoblastic proliferation activity. In current study, we investigated potential effects on activity, ROS...

Trabecular titanium implants are widely used in orthopedic surgery and known to promote osseointegration. In this study, we investigated whether primary human osteoblast-like cells grown inside a 3D trabecular scaffold undergo changes migration capacity, transcriptomic profile, cellular phenotype as compared the same osteoblasts not scaffold. Scratch tests have shown that of from external environment into Next generation sequencing analysis demonstrated modified expression genes involved...

The osteoprotegerin (OPG)/receptor activator of nuclear factor-κB ligand (RANKL)/receptor (RANK) system was evaluated as a potential target CGRP anabolic activity on bone. Primary cultures human osteoblast-like cells (hOB) express calcitonin receptor-like receptor (CLR) and modifying protein 1, and, because stimulates cAMP (one the modulators OPG production in osteoblasts), it investigated whether affects secretion expression hOB. treatment hOB (10 −11 M–10 −7 M) dose-dependently inhibited...
