Lysine methyltransferase KMT5C catalyzes deposition of trimethylation on histone H4 lysine 20 (H4K20me3), an epigenetic marker usually associated with gene repression and maintenance heterochromatin. is widely expressed in a variety tissues, however, its functional role liver has not been explored. Here, we show Kmt5c fasting- glucagon-induced which regulates hepatic gluconeogenesis. Loss hepatocytes results downregulated gluconeogenic expression compromised glucose output during fasting....

Abstract It is well known that c-Src has important roles in tumorigenesis. However, it remains unclear whether contributes to metabolic reprogramming. Here we find can interact with and phosphorylate hexokinases HK1 HK2, the rate-limiting enzymes glycolysis. Tyrosine phosphorylation dramatically increases their catalytic activity thus enhances Mechanistically, of at Tyr732 robustly decreases its K m V max by disrupting dimer formation. Brown and beige fat are specialized for energy expenditure by dissipating from glucose fatty acid oxidation as heat. While metabolism have been extensively studied in thermogenic adipose tissues, the involvement of amino acids regulating adaptive thermogenesis remains little studied. Here, we report that asparagine supplementation brown adipocytes drastically upregulated transcriptional program lipogenic gene expression, so asparagine-fed mice showed better cold tolerance. In with...

Mitochondria are the pivot organelles to control metabolism and energy homeostasis. The capacity of mitochondrial metabolic adaptions cold stress is essential for adipocyte thermogenesis. How brown adipocytes keep fitness upon a challenge cold-induced oxidative has not been well characterized. This manuscript shows that IFI27 plays an important role in cristae morphogenesis, keeping intact succinate dehydrogenase (SDH) function active fatty acid oxidation sustain thermogenesis adipocytes....

Two hallmarks of cancer cells are their resistance to apoptosis and ability thrive despite reduced levels vital serum components. c-jun N-terminal kinase (JNK) activation is crucial for triggered by starvation (SS), isocitrate dehydrogenase 1 (IDH1) mutations tumorigenic, in part, because they produce the abnormal metabolite 2-hydroxyglutarate (2-HG). However, it unknown whether 2-HG-induced tumorigenesis partially due JNK inhibition thus defective SS-induced apoptosis. We show here, using...

Significance The thermogenic function of adipocytes relies on the activation program that is coordinately modulated by an array unique transcriptional and epigenetic regulators. Histone methylations represent essential mode regulation in establishment tissue-specific gene expression. However, role H4K20 methylation not completely elucidated.
