We searched for germ-line mutations of the APC gene in 79 unrelated patients with familial adenomatous polyposis using a ribonuclease protection analysis coupled polymerase chain reaction amplifications genomic DNA. Mutations were found 53 (67%); 28 small deletions and 2 1- to 2-base-pair insertions; 19 point resulting stop codons only 4 missense mutations. Thus, 92% predicted result truncations protein. More than two-thirds (68%) clustered 5' half last exon, nearly two-fifths total occurred...
Recently, nonalcoholic steatohepatitis (NASH) was found to be correlated with cardiovascular disease events independently of the metabolic syndrome. The aim this study investigate whether an atherogenic (Ath) diet induces pathology necessary for diagnosis human NASH and how cholesterol triglyceride alter hepatic gene expression profiles responsible oxidative stress. We investigated liver plasma lipids mice fed Ath diet. profile examined microarrays real-time polymerase chain reactions....
Visceral adiposity in obesity causes excessive free fatty acid (FFA) flux into the liver via portal vein and may cause disease hepatic insulin resistance. However, because animal models of resistance induced by lipid infusion or a high fat diet are complex be accompanied alterations not restricted to liver, it is difficult determine contribution FFAs Therefore, we treated H4IIEC3 cells, rat hepatocyte cell line, with monounsaturated (oleate) saturated (palmitate) investigate direct initial...
A member of a novel family the human major histocompatibility complex (MHC) class I genes termed MIC (MHC chain-related genes), MICA, has been recently identified near HLA-B gene on short arm chromosome 6. The predicted amino acid sequence MICA chain suggests that it folds similarly to typical chains and may have capacity bind peptides or other ligands. Therefore, is specialized function in antigen presentation T cell recognition. During nucleotide analyses genomic clone, we found triplet...
Various peripheral tissues show circadian rhythmicity, which is generated at the cellular level by their own core oscillators that are composed of transcriptional/translational feedback loops involving a set clock genes. Although circulating levels some adipocytokines, i.e. bioactive substances secreted adipocytes, on 24-h rhythmic cycle, it remains to be elucidated whether gene system works in adipose tissue. To address this issue, we investigated daily mRNA expression profiles genes and...
Abstract Recent studies have demonstrated relationships between the dysfunction of circadian clocks and development metabolic abnormalities, but chicken-and-egg question remains unresolved. To address this issue, we investigated cause-effect relationship in obese, diabetic ob/ob mice. Compared with control C57BL/6J mice, daily mRNA expression profiles clock clock-controlled genes Clock, Bmal1, Cry1, Per1, Per2, Dbp were substantially dampened liver adipose tissue, not hypothalamic...
Background Optimal treatment for nonalcoholic steatohepatitis (NASH) has not yet been established, particularly individuals without diabetes. We examined the effects of metformin, commonly used to treat patients with type 2 diabetes, on liver pathology in a non-diabetic NASH mouse model. Methodology/Principal Findings Eight-week-old C57BL/6 mice were fed methionine- and choline-deficient plus high fat (MCD+HF) diet or 0.1% metformin 8 weeks. Co-administration significantly decreased fasting...
Human T-lymphotropic virus type 1 (HTLV-1) is linked to multiple diseases, including the neuroinflammatory disease HTLV-1–associated myelopathy/tropical spastic paraparesis (HAM/TSP) and adult T cell leukemia/lymphoma. Evidence suggests that HTLV-1, via viral protein Tax, exploits CD4+ plasticity induces transcriptional changes in infected cells cause suppressive CD4+CD25+CCR4+ Tregs lose expression of transcription factor FOXP3 produce IFN-γ, thus promoting inflammation. We hypothesized...
Development of severe sepsis is thought to result from the overproduction cytokines, such as tumor necrosis factor-alpha (TNF-alpha) and interleukin-1beta (IL-1beta), nitric oxide. Recently, 3-hydroxy-3-methylglutaryl coenzyme A reductase inhibitors, which are antihypercholesterolemic agents, have been reported inhibit lipopolysaccharide (LPS)-induced production cytokines oxide in vitro. In this study, we tested these effects vivo. After LPS administration (15 mg/kg i.p.) CD-1 mice, serum...
We recently identified a trinucleotide repeat polymorphism, (GCT)n, within the transmembrane (TM) segment of human MHC class I MICA gene (MHC chain‐related A). Five distinct alleles (A4, A5, A5.1, A6, A9) corresponding to 4, 5, 5 with one nucleotide insertion, 6 and 9 repetitions, respectively, have been detected in various HLA‐homozygous B cell lines. Here we present allele frequencies for this trimeric short tandem (STR) 604 unrelated individuals collected from nine populations (Japanese,...
Background Human T-lymphotropic virus type 1 (HTLV-1) -associated myelopathy/tropical spastic paraparesis (HAM/TSP) is a rare chronic neuroinflammatory disease. Since the disease course of HAM/TSP varies among patients, there dire need for biomarkers capable predicting rate progression. However, have been no studies to date that compared prognostic values multiple potential HAM/TSP. Methodology/Principal Findings Peripheral blood and cerebrospinal fluid (CSF) samples from patients...
Human T-lymphotropic virus type 1-associated myelopathy/tropical spastic paraparesis (HAM/TSP) is a rare neurodegenerative disease characterized by chronic inflammation in the spinal cord. We hypothesized that positive feedback loop driven chemokines may be responsible for HAM/TSP. aimed to determine identity of these chemokines, where they are produced, and how drive found patients with HAM/TSP have extraordinarily high levels chemokine CXCL10 (also known as IP-10) an abundance cells...
Meningiomas are among the most common intracranial tumors and mostly curable by surgical resection. However, some populations of meningiomas with benign histological profiles show malignant behavior. The reasons for this inconsistency yet to be ascertained, novel diagnostic criteria other than one urgently needed. aim present study is subclassify from viewpoint gene methylation determine subgroup characteristics. Thirty were analyzed using microarrays 6157 genes classified into three...
Drug-induced nephrotoxicity is a serious problem in patients with hospital-acquired acute kidney injury (AKI). A new renal biomarker needed because traditional markers are not sensitive for early detection of drug-induced AKI. In recent study, we demonstrated that vanin-1 novel candidate nephrotoxicant-induced injury. The objective the present study to determine whether increase urinary detected before elevations serum creatinine or N-acetyl-β-glucosaminidase (NAG), molecule-1...
Aims To compare the effects of grapefruit juice (GFJ) on pharmacokinetics pitavastatin and atorvastatin. Methods In a randomized, four‐phase crossover study, eight healthy subjects consumed either GFJ or water t.i.d. for 4 days in each trial. On final day, single dose mg 20 atorvastatin was administered. Results increased mean AUC 0−24 acid by 83% (95% CI 23–144%) that 13% (−3 to 29%). Conclusions Pitavastatin, unlike atorvastatin, appears be scarcely affected CYP3A4‐mediated metabolism.
