Acute graft-versus-host disease (GVHD) remains a major limitation of allogeneic stem-cell transplantation; not all patients have response to standard glucocorticoid treatment. In phase 2 trial, ruxolitinib, selective Janus kinase (JAK1 and JAK2) inhibitor, showed potential efficacy in with glucocorticoid-refractory acute GVHD.
PURPOSE Despite undergoing allogeneic hematopoietic stem cell transplantation (HCT), patients with acute myeloid leukemia (AML) internal tandem duplication mutation in the FMS-like tyrosine kinase 3 gene ( FLT3-ITD) have a poor prognosis, frequently relapse, and die as result of AML. It is currently unknown whether maintenance therapy using FLT3 inhibitors, such multitargeted inhibitor sorafenib, improves outcome after HCT. PATIENTS AND METHODS In randomized, placebo-controlled, double-blind...
Chronic graft-versus-host disease (GVHD), a major complication of allogeneic stem-cell transplantation, becomes glucocorticoid-refractory or glucocorticoid-dependent in approximately 50% patients. Robust data from phase 3 randomized studies evaluating second-line therapy for chronic GVHD are lacking. In retrospective surveys, ruxolitinib, Janus kinase (JAK1–JAK2) inhibitor, showed potential efficacy patients with -dependent GVHD.
Hepatocellular carcinoma (HCC) is the fifth most common malignancy worldwide with a poor prognosis and limited therapeutic options. To aid development of novel immunological interventions, we studied breadth, frequency, tumor-infiltration naturally occurring CD8+ T-cell responses targeting several tumor-associated antigens (TAA). We used overlapping peptides spanning entire alpha-fetoprotein (AFP), glypican-3 (GPC-3), melanoma-associated gene-A1 (MAGE-A1) New York-esophageal squamous cell...
The success of allogeneic hematopoietic cell transplantation is limited by acute graft-versus-host disease (GvHD), a severe complication accompanied high mortality rates. Yet, the molecular mechanisms initiating this remain poorly defined. In study, we show that, after conditioning therapy, intestinal commensal bacteria and damage-associated pattern uric acid contribute to Nlrp3 inflammasome–mediated IL-1β production that gastrointestinal decontamination depletion reduced GvHD severity....
Oncogenic JAK mutation sensitizes myeloproliferative neoplasms to immune checkpoint inhibition.
Abstract Mutational inactivation of VHL is the earliest genetic event in majority clear cell renal carcinomas (ccRCC), leading to accumulation HIF-1α and HIF-2α transcription factors. While correlative studies human ccRCC functional using lines have implicated as an inhibitor a promoter aggressive tumour behaviours, their roles onset not been functionally addressed. Herein we show autochthonous model that Hif1a essential for formation whereas Hif2a deletion has only minor effects on...
