Abstract Extracellular histones in neutrophil extracellular traps (NETs) or chromatin from injured tissues are highly pathological, particularly when liberated by DNases. We report the development of small polyanions (SPAs) (~0.9–1.4 kDa) that interact electrostatically with histones, neutralizing their pathological effects. In vitro, SPAs inhibited cytotoxic, platelet-activating and erythrocyte-damaging effects mechanistic studies revealing block disruption lipid-bilayers histones. vivo,...

Promoters comprise multiple elements to modulate transcription. Despite decades of research, significant sequence and length variation promoter renders elucidation function the evolutionary diversity transcriptional regulation challenging. Combining massively parallel assays with biophysical modeling, we mapped each element across 49 phylogenetically diverse bacterial genomes (GC%: 27.8-72.1). We discovered a conserved element, 'start,' that dictates transcription start site selection....
The first enantioselective total synthesis of (+)-(R)-concentricolide, the enantiomer an anti-HIV-1 agent isolated from Daldinia concentrica, 2-iodophenol in 7 steps reveals (S)-configuration for natural form furanophthalide. key features include anionic ortho-Fries rearrangement to furnish 3-iodosalicylamide, facile construction benzofuran system employing tandem Sonogashira coupling annulation reaction, directed ortho metalation introduce a propanoyl group, as well CBS reduction,...
This work describes an expeditious and efficient preparation of enantiopure (thiolan-2-yl)diphenylmethanol (2) featuring a double nucleophilic substitution Shi epoxidation as key steps. One the applications its benzyl ether derivative to asymmetric sulfur ylide-mediated with up 92% ee (14 examples) was also demonstrated herein.
The high affinity of highly charged polynuclear platinum complexes for glycans such as heparan sulfate results in modulation the biomolecule signaling functions leading to inhibition angiogenesis.
In this article, low-temperature Cu-Cu bonding with the Ag passivation layer was accomplished at 180 °C for 3 min without a postannealing process. The effectively prevents Cu from oxidation and assists diffusion to realize bonding. small grain size of controlled by sputtering working pressure is observed improve quality. Different plasma treatment parameters conditions were executed evaluate behavior investigated, corresponding mechanism discussed as well. addition, reliability assessments,...
Palladium(0)-catalyzed allylation of soft nucleophiles is a powerful synthetic methodology for the construction carboncarbon or carbon-heteroatom bonds. The possibility controlling regioselectivity one most interesting aspects this chemistry. In review, different factors that can affect regiochemistry nucleophilic attack will be discussed. Keywords: Palladium, allylic substitution, regioselectivity, nucleophiles, ligands, steric effects
Glucocorticoids are commonly used in treating diseases with white matter lesions, including demyelinating and spinal cord injury (SCI). However, glucocorticoids ineffective gray injuries, such as head stroke. The differential glucocorticoid effects injuries unclear. We report here a novel mechanism of methylprednisolone (MP), synthetic widely for multiple sclerosis SCI, protecting oligodendrocytes (OLGs) against AMPA-induced excitotoxicity, which has been implicated the diseases....
We report herein a detailed NMR study of the aquation and subsequent covalent binding trinuclear clinical agent [{trans-PtCl(15NH3)2}2{μ-trans-Pt(15NH3)2(15NH2(CH2)615NH2)2}]4+ (1, 1,0,1/t,t,t or Triplatin) with three d-glucosamine residues containing varied O-sulfate N-sulfate N-acetyl substitutions, which represent monosaccharide fragments present within repeating disaccharide sequences cell surface heparan sulfate (HS). The monosaccharides GlcNS(6S), GlcNS, GlcNAc(6S) were synthesized in...
Enterovirus 71 (EV71) is a major etiological agent of hand, foot, and mouth disease, for which there no antiviral therapy. We have developed densely sulfated disaccharide heparan sulfate (HS) analogues that are potent small molecule inhibitors EV71 infection, binding to the viral capsid acting as decoy receptors block early events virus replication. The simplified structures, more than defined HS disaccharides with significant anticoagulant activity, offer promise anti-EV71 agents.
A regio- and stereocontrolled solution for a selective modification at the C-2 or C-4 position of N-acetylneuraminic acid involves use allylic substitution catalyzed by palladium. As illustrated in scheme, regioselective malonylation is under strict influence ligands associated with allylpalladium complex.
Abstract Bacterial heparinases that cleave heparan sulfate (HS) and heparin are widely used to generate low‐molecular‐weight heparins (LMWHs) structurally functionally characterise HS biomolecules. We provide novel insights into the substrate specificity of heparinase II from two different bacteria: Pedobacter heparinus (formerly Flavobacterium heparinum ) Bacteroides eggerthii . The activity towards various well‐defined oligosaccharides was investigated by 1 H NMR spectroscopy; this...
Abstract A process for highly effective regio‐ and stereoselective palladium‐catalyzed allylic substitution of 2,3‐unsaturated derivatives N ‐acetylneuraminic acid (Neu5Ac2en) has been developed. We show that the efficiency allylation reaction depends on suitable protecting groups starting material that, with sodium malonate anion as a nucleophile, regioselectivity can be fine‐tuned by nature ligands associated palladium complex. These results are explained stoichiometric preparation study...
