Myelodysplastic syndromes (MDS) are heterogeneous myeloid neoplasms in which a risk-adapted treatment strategy is needed. Recently, new clinical-molecular prognostic model, the Molecular International Prognostic Scoring System (IPSS-M) was proposed to improve prediction of clinical outcome currently available tool (Revised [IPSS-R]). We aimed provide an extensive validation IPSS-M.
Synthetic data are artificial generated without including any real patient information by an algorithm trained to learn the characteristics of a source set and became widely used accelerate research in life sciences. We aimed (1) apply generative intelligence build synthetic different hematologic neoplasms; (2) develop validation framework assess fidelity privacy preservability; (3) test capability clinical/translational hematology.
Allogeneic hematopoietic stem-cell transplantation (HSCT) is the only potentially curative treatment for patients with myelodysplastic syndromes (MDS). Several issues must be considered when evaluating benefits and risks of HSCT MDS, timing being a crucial question. Here, we aimed to develop validate decision support system define optimal MDS on basis clinical genomic information as provided by Molecular International Prognostic Scoring System (IPSS-M).
Rare cancers constitute over 20% of human neoplasms, often affecting patients with unmet medical needs. The development effective classification and prognostication systems is crucial to improve the decision-making process drive innovative treatment strategies. We have created implemented MOSAIC, an artificial intelligence (AI)-based framework designed for multimodal analysis, classification, personalized prognostic assessment in rare cancers. Clinical validation was performed on...
Background: Papillary thyroid cancers (PTCs) are common, usually indolent malignancies. Still, a small but significant percentage of patients have aggressive tumors and develop distant metastases leading to death. Currently, it is not possible discriminate lesions due lack prognostic markers. Long noncoding RNAs (lncRNAs), which selectively expressed in context-dependent manner, expected represent new landscape search for molecular discriminants. Transforming growth factor β (TGFβ)...
Background: Growing evidence suggests that an altered microbiota composition contributes to the pathogenesis and clinical features in celiac disease (CD). We performed a comparative analysis of gut adulthood CD evaluate whether: (i) dysbiosis anticipates mucosal lesions, (ii) gluten-free diet restores eubiosis, (iii) refractory has peculiar microbial signature, (iv) salivary fecal communities overlap one. Methods: This is cross-sectional study where total 52 patients, including 13 active CD,...
Abstract The transcription factor NF-Y promotes cell proliferation and its activity often declines during differentiation through the regulation of NF-YA, DNA binding subunit complex. In stem compartments, shorter NF-YA splice variant is abundantly expressed sustains their expansion. Here, we report that satellite cells, population adult skeletal muscle necessary for growth regeneration, express uniquely longer isoform, majorly associated with differentiation. Through generation a...
Abstract Non-small-cell lung carcinoma (NSCLC) is the most common cancer and one of pioneer tumors in which immunotherapy has radically changed patients’ outcomes. However, several issues are emerging their implementation required to optimize immunotherapy-based protocols. In this work, we investigate ability Bromodomain Extra-Terminal protein inhibitors (BETi) stimulate a proficient anti-tumor immune response toward NSCLC. By using vitro, ex-vivo, vivo models, demonstrate that these...
Several computational pipelines for biomedical data have been proposed to stratify patients and predict their prognosis through survival analysis. However, these analyses are usually performed independently, without integrating the information derived from each of them. Clustering is an underexplored problem, current approaches limited applications, whose heterogeneous multimodal, with poor scalability high-dimensionality. We introduce VAE-Surv, a multimodal framework patients'...
Background Sex is a major source of diversity among patients and sex-informed approach becoming new paradigm in precision medicine. We aimed to describe sex myelodysplastic syndromes terms disease genotype, phenotype, clinical outcome. Moreover, we sought incorporate information into the decision-making process as fundamental component patient individuality. MethodsIn this multicentre, observational cohort study, retrospectively analysed 13 284 aged 18 years or older with diagnosis syndrome...
Abstract Tumor‐associated macrophages (TAMs) have a unique favorable effect on the prognosis of colorectal cancer (CRC), although their association with stage‐specific outcomes remains unclear. We assessed densities CD68 + and CD163 TAMs at invasive front resected CRC stage III from 236 patients, 165 whom received post‐surgical FOLFOX treatment, relationship disease‐free survival (DFS). Associations between macrophage mRNAs clinical outcome were investigated in silico 59 FOLFOX‐treated...
Abstract Chromatin modifiers are emerging as major determinants of many types cancers, including Anaplastic Large Cell Lymphomas (ALCL), a family highly heterogeneous T-cell lymphomas for which therapeutic options still limited. HELLS is multifunctional chromatin remodeling protein that affects genomic instability by participating in the DNA damage response. Although transcriptional function has been suggested, no clues on how controls transcription currently available. In this study,...
Decision about the optimal timing of a treatment procedure in patients with hematologic neoplasms is critical, especially for cellular therapies (most including allogeneic hematopoietic stem-cell transplantation [HSCT]). In absence evidence from randomized trials, real-world observational data become beneficial to study effect timing. this study, framework estimate expected outcome after an intervention time-to-event scenario developed, aim optimizing personalized manner.
Anaplastic Thyroid Cancer (ATC) is an undifferentiated and aggressive tumor that often originates from well-Differentiated Carcinoma (DTC) through a trans-differentiation process. Epithelial-to-Mesenchymal Transition (EMT) recognized as one of the major players this OVOL2 transcription factor (TF) promotes epithelial differentiation restrains EMT during embryonic development. loss in some types cancers linked to aggressiveness poor prognosis. Here, we aim clarify unexplored role ATC.Gene...
MicroRNAs, highly-conserved small RNAs, act as key regulators of many biological functions in both plants and animals by post-transcriptionally regulating gene expression through interactions with their target mRNAs. The microRNA research is a dynamic field, which new unconventional aspects are emerging alongside well-established roles development stress adaptation. A recent hypothesis states that miRNAs can be transferred from one species to another potentially genes across distant species....
Abstract Deregulation of chromatin modifiers, including DNA helicases, is emerging as one the mechanisms underlying transformation anaplastic lymphoma kinase negative (ALK − ) large cell (ALCL). We recently identified DNA-helicase HELLS central for proficient ALK ALCL proliferation and progression. Here we assessed in detail its function by performing RNA-sequencing profiling coupled with bioinformatic prediction to identify targets transcriptional cooperators. demonstrated that HELLS,...
Background: Endometrial cancer (EC) is the most common gynecologic tumor and world’s fourth in women. Most patients respond to first-line treatments have a low risk of recurrence, but refractory patients, those with metastatic at diagnosis, remain no treatment options. Drug repurposing aims discover new clinical indications for existing drugs known safety profiles. It provides ready-to-use therapeutic options highly aggressive tumors which standard protocols are ineffective, such as...
Long non-coding RNA (lncRNA) are emerging as powerful and versatile regulators of transcriptional programs distinctive biomarkers progression T-cell lymphoma. Their role in the aggressive anaplastic lymphoma kinase-negative (ALK–) subtype large cell (ALCL) has been elucidated only part. Starting from our previously identified ALCL-associated lncRNA signature performing digital gene expression profiling a retrospective cohort ALCL, we defined an 11 able to discriminate among ALCL subtypes. We...
