Background and Aims Lipopolysaccharides (LPS) is increased in nonalcoholic fatty liver disease (NAFLD), but its relationship with inflammation not defined. Approach Results We studied Escherichia coli LPS patients biopsy‐proven NAFLD, 25 simple steatosis (nonalcoholic liver) steatohepatitis (NASH), mice diet‐induced NASH. NASH had higher serum hepatocytes localization than controls, which was correlated zonulin phosphorylated nuclear factor‐κB expression. Toll‐like receptor 4 positive (TLR4...

The prevalence of cardiometabolic disorders, including non-alcoholic fatty liver disease (NAFLD), is increasing in western countries, because changes lifestyle and dietary habits. Mediterranean Diet (Med-Diet) effective for cardiovascular prevention, but its relationship with NAFLD has been scarcely investigated. We included 584 consecutive outpatients presenting one or more risk factor such as type 2 diabetes mellitus (T2DM), arterial hypertension, overweight/obesity, dyslipidemia. Liver...

Insulin resistance (IR) is the cornerstone of Metabolic Dysfunction Associated Steatotic Liver Disease (MASLD), pathophysiologically being key link between MASLD, metabolic disorders, and cardiovascular (CV) diseases. There are no prospective studies comparing predictive values different markers insulin in identifying presence MASLD associated risk events (CVEs).

Homozygous familial hypercholesterolemia (HoFH) is a rare genetic condition characterized by extremely increased low-density lipoprotein (LDL) cholesterol levels and premature atherosclerotic cardiovascular disease (ASCVD). Heterozygous (HeFH) more common than HoFH, women with HeFH are diagnosed later undertreated compared to men; it unknown whether these sex differences also apply HoFH.

Abstract Background Chronic oxidative stress is one of the key mechanisms responsible for disease progression in non-alcoholic fatty liver disease. However, so far, few studies reported increased circulating levels markers patients with and no study has been performed newer systemic stress. The aim was to assess relationship between urinary 8-iso-prostaglandin F2α serum soluble NOX2-derived peptide severity steatosis subjects liver. Methods 264 consecutive referred suspected metabolic...

Abstract Background Several studies suggest an increase of oxidative stress and a reduction endothelial function in obstructive sleep apnoea syndrome (OSAS). We assessed the association between OSAS, dysfunction stress. Further aim was to evaluate effect nasal continuous positive airway pressure (nCPAP) on arterial dysfunction. Methods studied 138 consecutive patients with heavy snoring possible OSAS. Patients underwent unattended overnight home polysomnography. Ten severe OSAS were...

NAFLD is a polygenic condition but the individual and cumulative contribution of identified genes remains to be established. To get additional insight into genetic architecture NAFLD, GWAS-identified GCKR, PPP1R3B, NCAN, LYPLAL1 TM6SF2 were resequenced by next generation sequencing in cohort 218 subjects 227 controls, where PNPLA3 rs738409 MBOAT7 rs641738 genotypes also obtained. A total 168 sequence variants detected 47 annotated as functional. When all functional within each gene...

Abstract Aims The availability of novel lipid-lowering therapies (LLTs) has remarkably changed the clinical management homozygous familial hypercholesterolaemia (HoFH). impact these advances was evaluated in a cohort 139 HoFH patients followed real-world setting. Methods and results characteristics patients, along with information about LLTs low-density lipoprotein cholesterol (LDL-C) levels at baseline after median follow-up 5 years, were retrospectively retrieved from records enrolled...

Non-alcoholic fatty liver disease (NAFLD) is characterized by intra-hepatic fat accumulation and mechanisms involved in its pathogenesis are not fully explained. Lysosomal Acid Lipase (LAL) a key enzyme lipid metabolism. We investigated activity patients with liver. LAL (nmol/spot/h) was measured 100 adult healthy subjects (HS) 240 NAFLD patients. A sub-analysis on 35 biopsy-proven non-alcoholic steatohepatitis (NASH) performed. Median 1.15 (0.95-1.72) HS. It significantly reduced [0.78...

Abstract Pathogenesis of non-alcoholic fatty liver disease (NAFLD) is influenced by predisposing genetic variations, dysmetabolism, systemic oxidative stress, and local cellular molecular cross-talks. Patatin-like phospholipase domain containing 3 (PNPLA3) gene I148M variant a known determinant NAFLD. Aims were to evaluate whether PNPLA3 was associated with specific histological pattern, hepatic stem/progenitor cell (HpSC) niche activation serum stress markers. Liver biopsies obtained from...

Oxidative stress plays a pivotal role in non-alcoholic fatty liver disease (NAFLD). Factors inducing oxidative NAFLD may be several; however, relationship with the adherence to Mediterranean Diet (Med-diet) and serum lipopolysaccharide (LPS) has been poorly investigated this setting. The aim was investigate factors associated impaired NAFLD, focusing on potential of LPS Med-diet. We enrolled 238 consecutive outpatients from PLINIO study, whom we measured soluble Nox2-derived peptide...

While it is known that circulating PCSK9 enhances platelet activation (PA) PCSK9i reduce it, the underlying mechanism not still clarified. Methods: In a multicenter before–after study in 80 heterozygous familial hypercholesterolemia (HeFH) patients on treatment with maximum tolerated statin dose ± ezetimibe, PA, soluble-NOX2-derived...
