The rice blast fungus Magnaporthe oryzae differentiates a specialized infection structure called an appressorium to invade cells. In this report, we show that CBP1, which encodes chitin-deacetylase, is involved in the induction phase of differentiation. We demonstrate enzymatic activity Cbp1 critical for formation. M. has six CDA homologues addition Cbp1, but none these are indispensable observed chitosan localization at fungal cell wall using OGA488. This observation suggests Cbp1-catalysed...

Clustered regularly interspaced short palindromic repeats (CRISPR)/CRISPR-associated protein 9 (Cas9)-mediated genome editing has become a promising approach for efficient and versatile genetic engineering in various organisms; however, simple precise nucleotide modification methods filamentous fungi have been restricted to double crossover type homologous recombination (HR). In this study, we developed novel strategy via single crossover-mediated HR the model fungus Pyricularia...

Mycoviruses are viruses that infect fungi. Unlike mammalian infectious viruses, their life cycle does not generally have an extracellular stage, and a symbiosis-like relationship is maintained between virus host Recently, mycoviruses been reported to show effects on fungi, altering biological properties such as growth rate, virulence, drug resistance, metabolite production. In this study, we systematically elucidated the of cells by comparing phenotypes transcriptomic responses in multiple...

RNA viruses in fungi (mycoviruses) are model systems for understanding the relationships between eukaryotic microorganisms and viruses. To reveal effects of mycoviruses on host fungi, it is essential to compare phenotypes isogenic fungal isolates with or without virus infection. Since active entry machinery has never been identified, introducing a difficult time-consuming process. Therefore, most studies have tried generate virus-free from infected strains by eliminating mycovirus. However,...

Abstract Chloramphenicol (Cm) is a broad-spectrum classic antibiotic active against prokaryotic organisms. However, Cm has severe side effects in eukaryotes of which the cause remains unknown. The plant pathogenic fungus Magnaporthe oryzae , causes rice blast, forms an appressorium to infect host cell via single-cell differentiation. specifically inhibits formation, indicates that novel molecular target (or targets) blast fungus. Application T7 phage display method inferred MoDullard,...

Metabolic switching and rewiring play a dynamic role in programmed cell differentiation. Many pathogenic microbes need to survive nutrient-deficient conditions use the glyoxylate cycle, an anaplerotic pathway of tricarboxylic acid produce carbohydrates. The plant fungus Magnaporthe oryzae (Pyricularia oryzae) has unique chitin deacetylase, Cbp1. spatiotemporal activity this protein is required for modification M. wall differentiation into specialized infection structure (appressorium). Here...

We report preliminary natural circular dichroism (NCD) spectra of amino acids, Land D-histidine, thin films in the nitrogen K-edge energy region. NCD peaks were observed at 399.9 eV and assigned to 1s → π* transition atoms imidazole ring. Imidazole ring is positioned side chain distant from asymmetric carbon. This result suggests that spectroscopy soft X-ray region can selectively pick up information on local structure around specific atoms, regardless position molecules. It interesting view...

Abstract The ascomycete Pyricularia oryzae (teleomorph: Magnaporthe oryzae) causes one of the most serious diseases known as rice blast. Nijmegen breakage syndrome protein (NBS1) is essential for DNA repair; thus, we studied P. NBS1 homolog (PoNBS1). A PoNBS1 null mutant exhibited high sensitivity to damage-inducing agents. also retarded hyphal growth, and induced abnormal conidial germination shape, but showed normal appressorium formation. phenotypes were complemented by introducing cDNA...
