Histone acetylation involves the transfer of two-carbon units to nucleus that are embedded in low-concentration metabolites. We found lactate, a high-concentration metabolic byproduct, can be major carbon source for histone through oxidation-dependent metabolism. Both cells and purified nuclei, 13C3-lactate carbons incorporated into H4 (maximum incorporation: ~60%). In nucleus, this process depends on nucleus-localized lactate dehydrogenase (LDHA), knockout (KO) which abrogates...

Although clinical and radiological examinations can be used to diagnose oral cancer, surgical pathology remains the gold standard, these conventional methods have limitations. We evaluated feasibility of longitudinal next-generation sequencing-based liquid biopsy for squamous cell carcinoma surveillance.Eleven patients were enrolled, plasma saliva collected before, 1, 3, 6 months after surgery. Tumor-specific mutations selected using paired, whole-exome analyses tumor tissues whole blood....

The Wnt and Hippo pathways are tightly coordinated understanding their reciprocal regulation may provide a novel therapeutic strategy for cancer. Anti-helminthic niclosamide is an effective inhibitor of now in phase II trial advanced colorectal cancer (CRC) patients. We found that Axin2, authentic target gene canonical Wnt, acts as aYAP phosphorylation activator APC-mutated CRC. While effectively suppresses it also inhibits Hippo, limiting its potential To overcome this limitation, we...

Dimorphic Candida exist as commensal yeast carriages or infiltrate hyphae in the oral cavity. Here, we investigated clinical relevance of non-pseudomembranous candidiasis (OC) by smears tongue biofilms. We conducted a retrospective study 2829 patients who had regardless OC suspicion. Clinical characteristics were evaluated using novel method assessing hyphae. factors (moderate/severe stimulated pain, pain aggravated stimulation, dorsum appearance and initial topical antifungal use) highly...

Tubulointerstitial renal fibrosis is a chronic disease process affecting kidney (CKD). While the etiological role of transforming growth factor-beta (TGF-β) well known for epithelial–mesenchymal transition (EMT) in disease, effective therapeutics are largely limited. As member TGF-β superfamily, bone morphogenetic protein-7 (BMP-7) plays an important as endogenous antagonist TGF-β, inhibiting fibrotic progression many organs. However, soluble rhBMP-7 hardly available due to its limited...

Background Approximately 30% of diabetic patients develop nephropathy, a representative microvascular complication. Although the etiological mechanism has not yet been fully elucidated, renal tubular damage by hyperglycemia-induced expression transforming growth factor-β (TGF-β) is known to be involved. Recently, new type cell death iron metabolism called ferroptosis was reported involved in kidney animal models which could induced TGF-β. Bone morphogenetic protein-7 (BMP7) well-known...

The concept of adequate surgical margins remains controversial in oral squamous cell carcinoma (OSCC) surgery. This study aimed to identify margin-related indicators that might impact recurrence and survival OSCC patients.Histopathological examination was performed using hematoxylin-eosin-stained margin tissue sections 235 patients. Cutibacterium acnes is a facultative anaerobic, gram-positive rod, and commensal bacterium of the body surface including oral cavity. A causal relationship between C. chronic granulomatous diseases, such as sarcoidosis orthopedic implant-associated infections, has been previously reported. Typically, observed inside macrophages, allowing evasion host immunity, triggering persistent inflammatory response. However, findings have not reported in peri-implantitis lesions. In this case series, we...

Background: Bone morphogenetic protein-7 (BMP-7) antagonises transforming growth factor-β (TGF-β). This study investigated the ability of a prodrug BMP-7, designed as micelle nanoparticles for nasal inhalation, to ameliorate pulmonary fibrosis in bleomycin (BLM)-induced murine model. Materials and Methods: Fluorescently labelled BMP-7 was delivered lungs via inhalation. Thirty-eight C57BL/6J mice were divided into three groups: control, BLM with BMP-7. We then administered vehicle nasally...

Schwannomas are benign tumors originating from myelinating cells constituting nerve sheaths but rarely contain cellular elements of the nerve. The authors encountered a 47-year-old female patient with schwannoma on anterior mandibular ramus arising buccal nerve, measuring 3 cm×4 cm. Surgical resection was performed preservation via microsurgical dissection. After one month, sensory function recovered without complications.
