ABSTRACT We evaluated the usefulness of matrix-assisted laser desorption ionization–time flight mass spectrometry (MALDI-TOF MS) for Cryptococcus identification at species and subspecies levels by using an in-house database 25 reference cryptococcal spectra. We report the first comparative evaluation between Bruker Biotyper MS (BMS) and Vitek (VMS) for identification of yeasts. The rate correct identifications at species level was comparable using commercial databases (89.8% versus 84.3%; P = 0.712), but higher BMS an in-house-extended database (100% 0.245). Importantly, misidentification significantly VMS (1% 12.1%; < 0.0001), including major errors (0% 4.5%; 0.0036).
