Programmed cell death-1 (PD-1)/programmed death ligand-1 (PD-L1) blocking therapy has become a major pillar of cancer immunotherapy. Compared with antibodies targeting, small-molecule checkpoint inhibitors which have favorable pharmacokinetics are urgently needed. Here we identified berberine (BBR), proven anti-inflammation drug, as negative regulator PD-L1 from set traditional Chinese medicine (TCM) chemical monomers. BBR enhanced the sensitivity tumour cells to co-cultured T-cells by...

The distal portion of rotavirus (RV) VP4 spike protein (VP8*) is implicated in binding to cellular receptors, thereby facilitating viral attachment and entry. While VP8* some animal RVs engage sialic acid, human often attach enter cells a acid-independent manner. A recent study demonstrated that the major (P[4], P[6], P[8]) recognize histo-blood group antigens (HBGAs). In this study, we performed phylogenetic analysis showed further variations RV interaction with HBGAs. On basis sequences,...

Abstract Hepatocellular carcinoma (HCC) is the third leading cause of cancer‐related death worldwide. The prognosis HCC remains very poor; thus, an effective treatment urgent. Herein, a type nanomedicine developed by conjugating Fe@Fe 3 O 4 nanoparticles with ginsenoside Rg3 (NpRg3), which achieves excellent coupling effect. In dimethylnitrosamine‐induced model, NpRg3 application significantly prolongs survival mice. Further research indicates that inhibits development and eliminates...

Abstract The acquisition of radioresistance by breast cancer cells during radiotherapy may lead to recurrence and poor survival. Signal transducer activator transcription 3 (Stat3) is activated in and, therefore, be an effective target for overcoming therapeutic resistance. Berberine (BBR), a traditional Chinese medicine, has therapeutic effects on variety of inflammation-related diseases, but its direct proteomic targets remain unknown. Using activity-based protein profiling, we first demonstrated that BBR directly the NEK7 via hydrogen bond between 2,3-methylenedioxy and 121-arginine (R121) residues. The fact R121 is located precisely within key domain involved in NEK7–NLRP3 interaction allows to specifically block successively inhibit IL-1β release,...
