Abstract A previous study demonstrated that a high-fat diet (HFD), administered for one-three-days, induces hypothalamic inflammation before obesity’s established, and the long term affects leptin signaling/action due to inflammation. We investigate whether exposure particulate matter of diameter ≤2.5 μm (PM 2.5 ) in mice fed with chow leads similar metabolic effects caused by feeding. Compared filtered air group (FA), one-day-exposure-PM did not affect adiposity. However,...

IKK epsilon (IKKε) is induced by the activation of nuclear factor-κB (NF-κB). Whole-body IKKε knockout mice on a high-fat diet (HFD) were protected from insulin resistance and showed altered energy balance. We demonstrate that expressed in neurons upregulated hypothalamus obese mice, contributing to leptin resistance. Blocking with CAYMAN10576 or small interfering RNA decreased NF-κB this tissue, relieving inflammatory environment. Inhibition activity, but not TBK1, reduced IRS-1Ser307...

Intestinal helminths are prevalent in individuals who live rural areas of developing countries, where obesity, type 2 diabetes, and metabolic syndrome rare. In the present study, we analyzed modulation gut microbiota mice infected with helminth Strongyloides venezuelensis, fed either a standard rodent chow diet or high-fat (HFD). To investigate effects on metabolism, expression tight-junction proteins epithelium, inflammatory markers serum tissue quantified glucose tolerance insulin...

Pulmonary arterial hypertension (PAH) is a vascular remodeling disease, characterized by increased blood pressure levels in pulmonary circulation, leading to restriction the circulation flow and heart failure. Although emergence of new PAH therapies has survival rates, this disease still high mortality patients that receive diagnosis die within few years. The pathogenesis involves multiple pathways, with complex interaction local distant cytokines, hormones, growth factors, transcription an...

Recent data show that iNOS has an essential role in ER stress obesity. However, whether is sufficient to account for obesity-induced and Unfolded Protein Response (UPR) not yet been investigated. In the present study, we used knockout mice investigate high-fat diet (HFD) can still induce residual stress-associated insulin resistance. For this purpose, intraperitoneal glucose tolerance test (GTT), euglycemic-hyperinsulinemic clamp, western blotting qPCR liver, muscle, adipose tissue of KO...

Abstract Extracellular vesicles (EVs) are small, lipid‐bilayer‐bound particles released by cells that can contain important bioactive molecules, including lipids, RNAs, and proteins. Once in the extracellular environment, EVs act as messengers locally well to distant tissues coordinate tissue homeostasis systemic responses. There is a growing interest not only understanding physiology of signaling but also leveraging them minimally invasive diagnostic prognostic biomarkers (e.g., they be...

Background Obesity results from imbalance between food intake and energy expenditure. The homeostasis is regulated by hypothalamic neurons that receive different neural, hormonal metabolic signals. Insulin one of the main hormones regulate acts through a cascade intracellular signaling depends on activation several proteins, such as Akt. Our hypothesis PH domain Leucine rich repeat Protein Phosphatase 1 (PHLPP1) inhibits Akt activity dephosphorylating serine 473 residues, impairing insulin...

Abstract Stress substantially increases the risk of developing painful temporomandibular disorders (TMDs) by influencing release endogenous catecholamines. Propranolol, an antagonist β‐adrenergic receptors, has shown potential in alleviating TMD‐associated pain, particularly when level catecholamines is elevated. The aim this study was to explore whether intra‐articular propranolol administration effective diminishing joint (TMJ) pain during repeated stress situations. Additionally, we...
