The increasing prevalence of antibacterial resistance globally underscores the urgent need to update antibiotics. Here, we describe a strategy for inducing self-assembly host-defense antimicrobial peptide (AMP) into nanoparticle antibiotics (termed nanobiotics) with significantly improved pharmacological properties. Our involves myristoylation human α-defensin 5 (HD5) as therapeutic target and subsequent in aqueous media absence exogenous excipients. Compared its parent HD5, C-terminally...

SignificanceWe report the development of peptidomimetic antibiotics derived from a natural antimicrobial peptide, human α-defensin 5. By engaging multiple bacterial targets, lead compound is efficacious in vitro and vivo against bacteria with highly inducible antibiotic resistance, promising useful therapeutic agent for treatment infections caused by antibiotic-resistant bacteria.

Abstract What We Already Know about This Topic Article Tells Us That Is New Background Pyroptosis, a type of proinflammatory programmed cell death, drives cytokine storm. Caspase-11–dependent macrophage pyroptosis contributes to mortality during sepsis. Sphingosine-1-phosphate receptor 2 (S1PR2) signaling can amplify interleukin-1β secretion in endotoxin-induced inflammation. Here, we hypothesized that S1PR2 increases caspase-11–dependent and worsens Gram-negative sepsis outcome. Methods A...

Background: Host defense peptides (HDPs) have emerged as a novel therapeutic paradigm for wound management; however, their clinical applications remain challenge owing to poor pharmacological properties and lack of suitable pharmaceutical formulations. Nanodefensin (ND), nanoengineered human α-defensin 5 (HD5), has shown improved relative the parent compound. In this study, we engineered nanodefensin-encased hydrogel (NDEFgel), investigated effects NDEFgel on healing, elucidated underlying...

Abstract Background: The World Health Organization (WHO) has endorsed school bystander cardiopulmonary resuscitation (CPR) training programs. But related researches in China are limited. Therefore, we conducted this study to assess CPR children and the impact of neighborhood socio-economic status (SES) on. Methods: A total 1,093 students from seven schools Zhejiang province participated study. Theoretical practical were instructor-led classes. Students completed a 10-statement questionnaire...

Endogenously produced glucocorticoids exhibit immunomodulating properties and are of pivotal importance for sepsis outcome. Uncontrolled activation the immune-adrenal crosstalk increases risk sepsis-related death. Triggering receptor expressed on myeloid cells-2 (TREM2) is richly macrophages has been demonstrated to improve outcome by enhancing elimination pathogens. However, role mode action macrophage TREM2 adrenocortical steroidogenesis remains unclear in septic shock. The acute shock...

Gram-negative sepsis has become a substantial and escalating global healthcare challenge due to the growing antibiotic resistance crisis sluggish development of new antibiotics. LL-37, unique Cathelicidin species found in humans, exhibits wide range bioactive properties, including direct bactericidal effects, inflammation regulation, LPS neutralization. Acute respiratory distress syndrome (ARDS) is a common lung disorder that involves severe inflammatory damage in the pulmonary barrier, but underlying mechanisms remain elusive. Here, we demonstrated macrophages originating from ARDS patients and mice caused by bacteria were characterized increased expression of ferroportin (FPN).
