Dynein-decorated doublet microtubules (DMTs) are critical components of the oscillatory molecular machine cilia, axoneme, and have luminal surfaces patterned periodically by microtubule inner proteins (MIPs). Here we present an atomic model 48-nm repeat a mammalian DMT, derived from cryoelectron microscopy (cryo-EM) map complex isolated bovine respiratory cilia. The structure uncovers principles organization features specific to vertebrate including previously unknown MIPs, bundle tektin...

Motile cilia defects impair cerebrospinal fluid (CSF) flow and can cause brain spine disorders. The development of ciliated cells, their impact on CSF flow, function in axial morphogenesis are not fully understood. We have characterized motile cells within the zebrafish ventricles. show that ventricles undergo restructuring through development, involving a transition from mono- to multiciliated (MCCs) driven by gmnc. MCCs co-exist with monociliated generate directional patterns. These...

A hallmark of meiosis is chromosomal pairing, which requires telomere tethering and rotation on the nuclear envelope through microtubules, driving chromosome homology searches. Telomere pulling toward centrosome forms "zygotene bouquet." Here, we identified cilium" in oocytes. This cilium provides a cable system for bouquet machinery extends throughout germline cyst. Using zebrafish mutants live manipulations, demonstrate that anchors to counterbalance pulling. The essential synaptonemal...

PURPOSE: To investigate if oxymatrine pretreatment could ameliorate renal I/R injury induced in rats and explore the possible role of Nrf2/HO-1 pathway. METHODS: Unilaterally nephrectomized were insulted by their left kidney. Twenty four randomly divided into three groups: sham group, + saline-treated OMT-treated group. Oxymatrine or vehicle solution was administered intraperitoneally injected 60 min before ischemia, respectively. Renal function, histology, makers oxidative stress, cell...

To investigate the effect of metformin on renal tubular epithelial cell apoptosis and inflammation after kidney ischemia/ reperfusion in rats.Eighteen SD rats were randomly divided into three groups: Sham (S), Ischemia/reperfusion (I/R), Metformin (E). Before establishing I/R model, group E was administered for days, while groups S equal volumes saline. After a right nephrectomy performed all groups, which left kidneys subjected to 45 min ischemia. Renal function, histology, assessed. AMPK,...

Classic endocytosis destinations include the recycling endosome returning to plasma membrane or late (LE) merging with lysosomes for cargo degradation. However, anti-angiogenic proteins angiostatin and isthmin, are endocytosed trafficked mitochondria (Mito) execute apoptosis of endothelial cells. How these extracellular reach remains a mystery. Through confocal super-resolution fluorescent microscopy, we demonstrate that isthmin through interaction between LE Mito. Using purified organelles,...

Patchoulol is a natural sesquiterpene alcohol with extensive applications in cosmetics and pharmaceuticals. In this study, we first constructed the synthesis pathway of patchoulol Saccharomyces cerevisiae by expressing synthase PTS gene using strong promoter GAL1. Afterward, metabolic flux precursor was enhanced strengthening mevalonate balancing competition pathway, resulting 32.74-fold increase production. Subsequently, supply acetyl-CoA yeast increased modifying transcriptional regulators...

Background: Hepatic ischemia-reperfusion injury (HIRI) stands as an unavoidable complication arising from liver surgery, profoundly intertwined with its prognosis. The role of lysine methyltransferase SET domain bifurcated 1 (SETDB1) in HIRI remains elusive, despite confirmation a potential therapeutic target for diverse diseases. Here, we investigated the mechanism by which SETDB1 regulated HIRI. Methods: RNA sequencing data were used to identify expression and targets through...

Numerous studies have demonstrated that microRNAs (miRNAs or miRs) play an important role in regulating osteogenic differentiation, but their specific regulatory mechanism requires further investigation. In the present study, it was revealed during differentiation of rat bone marrow mesenchymal stem cells (BMSCs), expression level miR-144-3p decreased with increased induction duration and negatively associated marker gene expression. Overexpression inhibited while inhibition promoted...

In this paper, a new method is proposed. left and right images their corresponding maps are used to render intermediate image respectively. Then two resulted from references fused, remained holes interpolated with neighbouring pixels holes. Experimental results show that proposed can achieve high object quality as well PSNR image.
