Doubled haploid technology has been widely applied to multiple plant species and is recognized as one of the most important technologies for improving crop breeding efficiency. Although mutations in MATRILINEAL/Zea mays PHOSPHOLIPASE A1/NOT LIKE DAD (MTL/ZmPLA1/NLD) Zea DOMAIN OF UNKNOWN FUNCTION 679 MEMBRANE PROTEIN (ZmDMP) have shown generate haploids maize, knowledge genetic basis induction (HI) remains incomplete. Therefore, cloning new genes underlying HI further elucidating its...

Abstract Tumor suppressor p53 (TP53) is frequently mutated in cancer, often resulting not only loss of its tumor-suppressive function but also acquisition dominant-negative and even oncogenic gain-of-function traits. While wild-type levels are tightly regulated, mutants typically stabilized tumors, which crucial for their properties. Here, we systematically profiled the factors that regulate protein stability mutant using marker-based genome-wide CRISPR screens. Most regulators mutants,...

Photothermal therapy and immunotherapy are emerging strategies for treating tumors. The combination of these two therapies can effectively leverage their advantages, thereby enhancing the therapeutic efficacy. Based on this premise, natural polysaccharide was prepared from edible Rosa roxburghii fruit applied to coat gold nanorods (GNRs). antitumor nanorods, designated as RRP-MPBA-GNRs, were developed combine photothermal immunotherapy. This system employed 4-mercaptophenylboronic acid...

This work aimed to investigate the effect of fucoidan (FPS) on urate transporters induced by uric acid (UA). The results showed that UA stimulated expression glucose transporter 9 (GLUT9) and 1 (URAT1) in HK-2 cells, FPS could reverse effect. Moreover, activate NF-κB, JNK PI3K/Akt pathways, but both pathway inhibitors inhibited UA-induced activation these three pathways. These data suggested effectively induction reabsorption URAT1 GLUT9 UA, through repressing signal pathways cells. vitro...

ABSTRACT Kaposi's sarcoma‐associated herpesvirus (KSHV) is the etiologic agent of several human cancers, including primary effusion lymphoma (PEL), usually seen in immunocompromised patients while lack effective therapeutic options. Interleukin‐1 (IL‐1) family a major mediator for inflammatory responses and has functional role both innate adaptive immunity. We previously showed high activation multiple IL‐1 signaling molecules during KSHV latent lytic stages, as well clinical samples from...

Ferrets have become an indispensable tool in the understanding of influenza virus virulence and pathogenesis. Furthermore, ferrets are preferred preclinical model for vaccine therapeutic testing. Here we characterized infectome during different stages infectious process with without prior specific immunity to influenza. RNA from lung tissue lymph nodes infected naïve animals was subjected next-generation sequencing, followed by de novo data assembly annotation resulting sequences; this...

Changes in the amount of mitochondrial DNA (mtDNA) have never been investigated plant zygotes or early embryos due to difficulty isolating these cells, although such changes mammalian embryos. Using single-cell quantitative real-time polymerase chain reaction (PCR) and laser confocal microscopy, we surveyed mtDNA levels during embryogenesis Torenia fournieri Arabidopsis thaliana. In contrast with embryos, which does not change, found that doubling occurred zygotic development T. two-cell...

Seed number and harvesting ability in maize (Zea mays L.) are primarily determined by the architecture of female inflorescence, namely ear. Therefore, ear morphogenesis contributes to grain yield as such is one key target traits during breeding. However, molecular networks this highly dynamic complex grain-bearing inflorescence remain largely unclear. As a first step toward characterizing these networks, we performed high-spatio-temporal-resolution investigation transcriptomes using 130...

The myotubularins are a family of phosphatases that dephosphorylate the phosphatidylinositols phosphatidylinositol-3-phosphate and phosphatidylinositol-3,5-phosphate. Several members mutated in disease, yet biological functions majority remain unknown. To gain insight into roles individual enzymes, we have used affinity purification coupled to mass spectrometry identify protein-protein interactions for myotubularins. myotubularin interactome comprises 66 high confidence (false discovery rate...
