Streptomyces sp. GF3587 and 3546 were found to be imine-reducing strains with high R- S-selectivity by screening using 2-methyl-1-pyrroline (2-MPN). Their whole-cell catalysts produced 91 mM R-2-methylpyrrolidine (R-2-MP) 99.2%e.e. 27.5 S-2-MP (92.3%e.e.) from 2-MPN at 91-92% conversion in the presence of glucose, respectively.

The (R)-imine reductase (RIR) of Streptomyces sp. GF3587 was purified and characterized. It found to be a NADPH-dependent enzyme, homodimer consisting 32 kDa subunits. Enzymatic reduction 10 mM 2-methyl-1-pyrroline (2-MPN) resulted in the formation 9.8 (R)-2-methylpyrrolidine ((R)-2-MP) with 99% e.e. enzyme showed not only activity for 2-MPN at neutral pH (6.5-8.0), but also oxidation (R)-2-MP under alkaline (10-11.5) conditions. appeared sulfhydryl based on sensitivity specific inhibitors....

Phosphatidylinositol binding clathrin assembly protein (PICALM), also known as lymphoid myeloid leukemia (CALM), was originally isolated part of the fusion gene CALM/AF10, which results from chromosomal translocation t(10;11)(p13;q14). CALM is sufficient to drive in vitro on lipid monolayers and regulates clathrin-coated budding size shape vesicles at plasma membrane. However, physiological role has yet be elucidated. Here, vivo investigated using CALM-deficient mice. mice exhibited retarded...

Brigatinib-based therapy was effective against osimertinib-resistant EGFR C797S mutants and is undergoing clinical studies. However, tumor relapse suggests additional resistance mutations might emerge. Here, we first demonstrated the binding mode of brigatinib to EGFR-T790M/C797S mutant by crystal structure analysis predicted brigatinib-resistant through a cell-based assay including N-ethyl-N-nitrosourea (ENU) mutagenesis. We found that clinically reported L718 G796 compound appeared,...

Abstract Mutated receptor tyrosine kinases (MT-RTKs) such as internal tandem duplication of FMS-like kinase 3 (FLT3 ITD) and a point mutation KIT D816V are driver mutations for acute myeloid leukemia (AML). Clathrin assembly lymphoid protein (CALM) regulates intracellular transport RTKs, however, the precise role MT-RTKs remains elusive. We here show that CALM knock down leads to severely impaired FLT3 ITD- or D814V-dependent cell growth compared marginal influence on wild-type FLT3-...
