Streptomyces sp. GF3587 and 3546 were found to be imine-reducing strains with high R- S-selectivity by screening using 2-methyl-1-pyrroline (2-MPN). Their whole-cell catalysts produced 91 mM R-2-methylpyrrolidine (R-2-MP) 99.2%e.e. 27.5 S-2-MP (92.3%e.e.) from 2-MPN at 91-92% conversion in the presence of glucose, respectively.
The (R)-imine reductase (RIR) of Streptomyces sp. GF3587 was purified and characterized. It found to be a NADPH-dependent enzyme, homodimer consisting 32 kDa subunits. Enzymatic reduction 10 mM 2-methyl-1-pyrroline (2-MPN) resulted in the formation 9.8 (R)-2-methylpyrrolidine ((R)-2-MP) with 99% e.e. enzyme showed not only activity for 2-MPN at neutral pH (6.5-8.0), but also oxidation (R)-2-MP under alkaline (10-11.5) conditions. appeared sulfhydryl based on sensitivity specific inhibitors....
Phosphatidylinositol binding clathrin assembly protein (PICALM), also known as lymphoid myeloid leukemia (CALM), was originally isolated part of the fusion gene CALM/AF10, which results from chromosomal translocation t(10;11)(p13;q14). CALM is sufficient to drive in vitro on lipid monolayers and regulates clathrin-coated budding size shape vesicles at plasma membrane. However, physiological role has yet be elucidated. Here, vivo investigated using CALM-deficient mice. mice exhibited retarded...
Brigatinib-based therapy was effective against osimertinib-resistant EGFR C797S mutants and is undergoing clinical studies. However, tumor relapse suggests additional resistance mutations might emerge. Here, we first demonstrated the binding mode of brigatinib to EGFR-T790M/C797S mutant by crystal structure analysis predicted brigatinib-resistant through a cell-based assay including N-ethyl-N-nitrosourea (ENU) mutagenesis. We found that clinically reported L718 G796 compound appeared,...
Tea intake has been associated with health benefits, including potential beneficial effects of catechin-containing teas on allergic symptoms. However, large-scale epidemiological studies the relationship between tea and symptoms have limited. The present study aimed to examine frequency cedar pollen allergy, which is a major cause seasonal hay fever in Japan, large Japanese cohort. Data antibody levels assessed by blood tests (green tea, coarse oolong black tea) self-administered...
A 30-year-old woman presented with infertility and irregular menstruation. Physical examination revealed obesity slightly pale conjunctivae. Transvaginal ultrasound polycystic ovary, bilateral hydrosalpinx, endometrial thickening. Magnetic resonance imaging of the pelvis showed several large cystic lesions elongated fallopian tubes. An biopsy was performed to investigate Pathological findings included small non-caseating epithelioid cell granulomas scattered plasma infiltration. The...
B cells play a central role in immune system function. Deregulation of normal cell maturation can lead to the development autoimmune syndromes as well malignancies. Elucidation molecular features is important for new target therapies diseases and Employing cell-specific conditional knockout mice, we have demonstrated here that transcription factor leukemia/lymphoma-related (LRF) forms an obligate dimer regulates mature lineage fate humoral responses via distinctive mechanisms. Moreover, LRF...
The reduction of skin blood flow induced by local cooling results from a reflex increase in sympathetic output and an enhanced vasoconstrictor activity cutaneous vessels. present study investigated the latter response vivo tetrodotoxin-treated mice, which nerve tone was abolished.Male ddY anaesthetized with pentobarbitone, were treated tetrodotoxin artificially ventilated. plantar (PSBF) measured laser Doppler flowmetry.Cooling air temperature around left foot 25 to 10 degrees C decreased...
Abstract Mutated receptor tyrosine kinases (MT-RTKs) such as internal tandem duplication of FMS-like kinase 3 (FLT3 ITD) and a point mutation KIT D816V are driver mutations for acute myeloid leukemia (AML). Clathrin assembly lymphoid protein (CALM) regulates intracellular transport RTKs, however, the precise role MT-RTKs remains elusive. We here show that CALM knock down leads to severely impaired FLT3 ITD- or D814V-dependent cell growth compared marginal influence on wild-type FLT3-...
Abstract Objectives Comorbidities are associated with a high burden of disease in people living HIV (PLWH). The objective was to investigate the prevalence chronic comorbidities and use co‐medications PLWH Japan. Methods This study retrospectively analysed clinical information from receiving antiretroviral therapy (ART) between April 2009 March 2019. Demographic characteristics, numbers types comorbidities, non‐ART co‐medications, were described by age groups. source data National Database...
Abstract To report the status of switch rates and time-to-switch antiretroviral therapy (ART) regimens by evaluating anchor drug classes common switching patterns in Japanese people living with human immunodeficiency virus (HIV, PLWH). This cross-sectional cohort study extracted data 28,089 PLWH from National Database Health Insurance Claims Specific Checkups Japan (NDB), which contains representing entire population Japan. first prescription records ART administered between January 2011...
Abstract Epigenetic regulation is required to ensure the precise spatial and temporal pattern of gene expression that necessary for embryonic development. Although roles some epigenetic modifications in development have been investigated depth, role methylation at lysine 79 (H3K79me) poorly understood. Dot1L, a unique methyltransferase H3K79, forms complexes with distinct sets co-factors. To further understand H3K79me embryogenesis, we generated mouse knockout Mllt10 , encoding Af10, one...
Pneumonia is the third most common cause of death in Japan. Low vaccination rates are thought to be related low levels public subsidy. Since 2014, Japanese government has offered subsidies through a 5-year national routine program 23-valent pneumococcal polysaccharide vaccine (PPV23) for older adults at age ≥65 years with intervals. We previously reported that, 2 into program, estimated rate was 40.6% end 2015, significant increase compared periods before introduced. Here, we present an...
High expression of thymidylate synthase (TS) and inactivation p53 are allegedly associated with chemoresistance. The authors evaluated TS in gastric cancer treated neoadjuvant S-1/cisplatin chemotherapy. Paraffin sections pretreatment biopsy surgical specimens from 41 cancers were immunostained for protein after appropriate antigen retrieval. Fifty-one cases without chemotherapy also studied. In the biopsies, high was seen 8% histologic responders, 28% nonresponders 31% controls. observed...
Here, we address the function of protein phosphatase 6 (PP6) loss on K‐ras‐initiated tumorigenesis in keratinocytes. To do so, developed tamoxifen‐inducible double mutant ( K‐ras G12D ‐expressing and Ppp6c ‐deficient) mice which expression is driven by cytokeratin 14 (K14) promoter. Doubly‐mutant showed early onset tumor formation lips, nipples, external genitalia, anus palms, had to be killed 3 weeks after induction tamoxifen, while comparably‐treated did not. H&E‐staining lip tumors...
In this study, we compared Ma Huang Tang (maoto), a traditional Japanese medicine (Kampo), with antiviral drugs to evaluate their respective and combined effect on the duration of fever other subjective symptoms influenza. Forty-five patients enrolled in randomized control trial had positive type A influenza rapid antigen test, provided written consent, sought treatment at Juntendo University Hospital between November 2008 March 2009. Using computer-gen- erated list, were randomly assigned...
CALM is implicated in the formation of clathrin-coated vesicles, which mediate endocytosis and intracellular trafficking growth factor receptors nutrients. We previously found that CALM-deficient mice suffer from severe anemia due to impaired clathrin-mediated transferrin receptor immature erythroblast. However, has been supposed regulate survival hematopoietic stem/progenitor cells. So, this study, we focused on function these here show number Linage−Sca-1+KIT+ (LSK) cells decreased fetal...
