Abstract Background Low-density lipoprotein cholesterol (LDL-C) is suggested to play a role in osteoporosis but its association with bone metabolism remains unclear. Effects of LDL-C-lowering drugs on are also controversial. We aim determine whether LDL-C linked causally mineral density (BMD) and assess the effects BMD. Methods Association between blood lipid levels BMD was examined by epidemiological observation analyses US representative cohort NHANES III (n = 3638) Hong Kong Osteoporosis...

Osteoporosis, a disease characterized by low bone mineral density (BMD), increases the risk for fractures. Conventional factors alone do not completely explain measured BMD or osteoporotic fracture risk. Metabolomics may provide additional information. Severe COVID-19 has a poor prognosis, while the genetic mechanism underlying severe remains largely unknown. We aimed to identify genes that are potentially causally associated with COVID-19. conducted summary data-based Mendelian randomization (SMR) analysis using expression quantitative trait loci (eQTL) data from 49 different tissues as exposure and three COVID-19-phenotypes (very respiratory confirmed [severe COVID-19], hospitalized COVID-19, SARS-CoV-2 infection) outcomes. The size of the human head is highly heritable, but genetic drivers its variation within general population remain unmapped. We perform a genome-wide association study on (N = 80,890) and identify 67 loci, which 50 are novel. Neuroimaging studies show that 17 variants affect specific brain areas, most have widespread effects. Gene set enrichment observed for various cancers p53, Wnt, ErbB signaling pathways. Growth differentiation factor 15 (GDF15) is a key regulator of body weight in animals by regulating food intake. Its receptor, glial cell-derived neurotrophic receptor alpha-like (GFRAL), was identified recently. Pre-clinical studies showed that it promising therapeutic target for cardiometabolic diseases and anorexia/cachexia. Although many pharmaceutical companies are developing drugs targeting GFRAL, whether the findings from animal can be extrapolated to man unknown. Mendelian...
