Many individuals with cancer are resistant to immunotherapies. Here, we identify the gene encoding pyrimidine salvage pathway enzyme cytidine deaminase (CDA) among top upregulated metabolic genes in several immunotherapy-resistant tumors. We show that CDA cells contributes uridine diphosphate (UDP) pool. Extracellular UDP hijacks immunosuppressive tumor-associated macrophages (TAMs) through its receptor P2Y
Severe corneal damage leads to complete vision loss, thereby affecting life quality and impinging heavily on the healthcare system. Current clinical approaches manage wounds suffer from severe drawbacks, thus requiring development of alternative strategies. Of late, mesenchymal stromal/stem cell (MSC)-derived extracellular vesicles (EVs) have become a promising tool in ophthalmic field. In present study, we topically delivered bone-marrow-derived MSC-EVs (BMSC-EVs), embedded methylcellulose,...
Abstract The p140Cap adaptor protein is a tumor suppressor in breast cancer associated with favorable prognosis. Here we highlight function of orchestrating local and systemic tumor-extrinsic events that eventually result inhibition the polymorphonuclear myeloid-derived cell creating an immunosuppressive tumor-promoting environment primary tumor, premetastatic niches at distant sites. Integrative transcriptomic preclinical studies unravel controls epistatic axis where, through upstream...
Aims: Biliary diseases represent around 10% of all chronic liver and affect both adults children. Currently available biochemical tests detect cholestasis but not early fibrosis. Circulating extracellular vesicles (EVs) provide a noninvasive, real-time molecular snapshot the injured organ. We thus aimed at searching for panel EV-based biomarkers cholestasis-induced fibrosis using mouse models. Results: Progressive detectable histological evidence collagen deposition was observed from day 8...
The RNA-binding protein, Epithelial Splicing Regulatory Protein 1 (ESRP1) can promote or suppress tumorigenesis depending on the cell type and disease context. In colorectal cancer, we have previously shown that aberrantly high ESRP1 expression drive tumor progression. order to unveil mechanisms by which modulate cancer traits, searched for proteins affected modulation of Esrp1 in two human lines, HCA24 COLO320DM, proteomics analysis. Proteins hosted endogenous ribonucleoprotein complex...
RNA binding proteins are well recognized as critical regulators of tumorigenic processes through their capacity to modulate biogenesis, including alternative splicing, stability and mRNA translation. The protein Epithelial Splicing Regulatory Protein 1 (ESRP1) can act a tumor suppressor or promoter in cell type- disease context-dependent manner. We have previously shown that elevated expression ESRP1 colorectal cancer cells drive progression. To gain further insights into the pro-tumorigenic...
Feline leukemia virus C receptor 1a (FLVCR1a), initially identified as a retroviral and localized on the plasma membrane, has emerged crucial regulator of heme homeostasis. Functioning positive δ-aminolevulinic acid synthase 1 (ALAS1), rate-limiting enzyme in biosynthetic pathway, FLVCR1a influences TCA cycle cataplerosis, thus impacting flux interconnected metabolic pathways. This study reveals an unexplored link between FLVCR1a, synthesis, cholesterol production endothelial cells. Using...
