Two coding variants of apolipoprotein L1 (APOL1) called G1 and G2 explain much the excess risk kidney disease in African Americans. While various cytotoxic phenotypes have been reported experimental models, proximal mechanism by which cause is poorly understood. Here, we leveraged three models a recently small molecule blocker APOL1 protein, VX-147, to identify upstream G1-induced cytotoxicity. In HEK293 cells, demonstrated that G1-mediated Na+ import/K+ efflux triggered activation G...

Resveratrol, a natural polyphenol compound contained in numerous plants, has been proposed as treatment for obesity-related disease processes such insulin resistance. However, humans there are conflicting results concerning the efficacy of resveratrol improving action; purpose present study was to determine whether obesity status (lean, severely obese) affects response human skeletal muscle. Primary muscle cells were derived from biopsies obtained age-matched lean and insulin-resistant women...

Skeletal muscle insulin resistance is a hallmark of Type 2 diabetes (T2DM) and may be exacerbated by protein modifications methylglyoxal (MG), known as dicarbonyl stress. The glyoxalase enzyme system composed 1/2 (GLO1/GLO2) the natural defense against stress, yet its expression, activity, regulation remain largely unexplored in skeletal muscle. Therefore, this study investigated stress subjects with T2DM (age: 56 ± 5 yr.; BMI: 32 kg/m ) compared lean healthy control (LHC; age: 27 1 22 )....

Exercise/exercise training can enhance insulin sensitivity through adaptations in skeletal muscle, the primary site of insulin-mediated glucose disposal; however, humans range improvement vary substantially. The purpose this study was to determine if obesity influences magnitude exercise response relation improving human muscle. Electrical pulse stimulation (EPS; 24 h) muscle myotubes improved action tissue from both lean and severely obese individuals, but responses EPS were blunted with...

Abstract Context Recent preclinical data suggest exercise during pregnancy can improve the metabolic phenotype not only of mother, but developing offspring as well. However, investigations in human are lacking. Objective To characterize effect maternal aerobic on offspring’s mesenchymal stem cells (MSCs). Design Randomized controlled trial. Setting Clinical research facility. Patients Healthy female adults between 18 and 35 years age ≤ 16 weeks’ gestation. Intervention Mothers were...

This study is the first to investigate effects of aerobic exercise intensity on circulating sRAGE isoforms, muscle RAGE protein, and splicing. isoforms tended diminish with exercise, although this effect was attenuated increasing intensity. Muscle protein gene expression were unaffected by exercise. However, individuals obesity displayed nearly twofold higher expression, which positively correlated P65 subunit NF-κB.

The progression of metabolic-dysfunction-associated steatotic liver disease (MASLD) to steatohepatitis (MASH) involves complex alterations in both liver-autonomous and systemic metabolism that influence the liver's balance fat accretion disposal. Here, we quantify relative contribution hepatic oxidative pathways injury MASLD-MASH. Using NMR spectroscopy, UHPLC-MS, GC-MS, performed stable-isotope tracing formal flux modeling fluxes humans across spectrum MASLD-MASH, mouse models impaired...

Thioredoxin-interacting protein (TXNIP) negatively effects the redox state and growth signaling via its interactions with thioredoxin (TRX) regulated in development DNA damage response 1 (REDD1), respectively. TXNIP expression is downregulated by pathways activated during aerobic exercise (AE), posttranslational modifications (PTMs; serine phosphorylation ubiquitination). The purpose of this investigation was to determine acute AE on expression, modifications, interacting partners, REDD1...

Preclinical rodent and nonhuman primate models investigating maternal obesity have highlighted the importance of intrauterine environment in development insulin resistance offspring; however, it remains unclear if these findings can be translated to humans. To investigate possible effects humans, we isolated mesenchymal stem cells (MSCs) from umbilical cord tissue infants born mothers normal weight or with obesity. Insulin-stimulated glycogen storage was determined MSCs undergoing myogenesis...

Objective To determine the effects of aerobic on metabolic biomarkers pregnancy, with expected lower levels glucose, insulin, lipids among exercise‐trained pregnant women. Methods Secondary data analyses were performed using from two, longitudinal prenatal...

Recent evidence identifies a potent role for aerobic exercise to modulate the activity of hypothalamic neurons related appetite; however, these studies have been primarily performed in male rodents. Since females markedly different neuronal mechanisms regulating food intake, current study aimed determine effects acute treadmill on neuron populations involved appetite female mice. Emerging evidence identifies a potent role for aerobic exercise to modulate activity of neurons involved in regulating appetite; however, these studies produce conflicting results. These discrepancies may be, part, due methodological differences, including differences intensity and pre-exercise energy status. Consequently, the current study utilized translational, well-controlled, within-subject, treadmill protocol investigate differential effects status on post-exercise feeding behavior...
