旨在探究HMGA1基因在猪不同组织、不同日龄下不同品种中的表达谱,并对该基因编码区及3'UTR区的多态性与猪背膘厚进行关联分析。本研究利用实时荧光定量PCR检测大白猪HMGA1基因在7种组织的表达情况,及其在60、150和210日龄大白猪和民猪背部脂肪中的差异表达。对576头大白猪×民猪F2代资源群体的基因组DNA重测序进行HMGA1序列分析,筛选其编码区及3'UTR区SNPs,并以屠宰体重为协变量与背膘厚(第6~7肋)进行关联分析。结果表明,HMGA1 mRNA在大白猪不同组织中均有表达,且在组织间存在显著差异(P 0.05)。通过对F2群体HMGA1序列分析,在编码区检测出13个单核苷酸多态性(single nucleotide polymorphism,SNP)位点,其中包含2个错义突变位点:g.3261G>A和g.5818G>A,且g.3261G>A位点与背膘厚显著关联(P C和g.7280T>C与背膘厚显著关联(P A、g.7217G>C和g.7280T>C位点可作为猪背膘厚选育的潜在分子标记,为猪的分子育种奠定基础。
