Limited data are available on breakthrough COVID-19 in patients with hematologic malignancy (HM) after anti-severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) vaccination. Adult HM, ≥1 dose of anti-SARS-CoV-2 vaccine, and between January 2021 March 2022 were analyzed. A total 1548 cases included, mainly lymphoid malignancies (1181 cases, 76%). After viral sequencing 753 (49%), the Omicron variant was prevalent (517, 68.7%). Most received ≤2 vaccine doses before (1419, 91%), mostly...

Abstract Chimeric Antigen Receptor T cells (CAR-T) are an outbreaking treatment option for relapsed/refractory (R/R) diffuse large B-cell lymphoma (DLBCL). Cytokine release syndrome (CRS) and immune effector cell-associated neurotoxicity (ICANS) the most common specific toxicities, while severe neutropenia infections often observed as well. From March 2020, early G-CSF prophylaxis at day (D) two post-infusion was systematically proposed. We then compared patients treated before that date who...

Abstract Patients with previous CD19-directed chimeric antigen receptor (CAR) T-cell therapy have a prolonged vulnerability to viral infections. Coronavirus disease 2019 (COVID-19) has great impact and previously been shown cause high mortality in this population. Until now, real-world data on the of vaccination treatment patients COVID-19 after CAR are lacking. Therefore, multicenter, retrospective study was conducted from EPICOVIDEHA survey. Sixty-four were identified. The overall caused...

Extranodal NK/T-cell lymphoma (ENKTCL) is an Epstein-Barr virus (EBV)-related neoplasm with male dominance and a poor prognosis. A better understanding of the genetic alterations their functional roles in ENKTCL could help improve patient stratification treatments. In this study, we performed comprehensive analysis 178 cases to delineate landscape mutations, copy number (CNA), structural variations, identifying 34 driver genes including six previously unappreciated ones, namely, HLA-B,...

Classical Hodgkin Lymphoma incidence increases in HIV-1-infected patients (HIV-cHL). HIV infection is associated with higher B-cell activation. Here, 38 HIV-cHL from the French cohort ANRS-CO16 Lymphovir, we examined longitudinally over 24 months serum levels of activating cytokines IL10, IL6, and BAFF, blood distribution subsets. Fourteen were also compared to matched HIV-infected controls without cHL. BAFF than (p < 0.0001, p = 0.002, respectively). Cytokine increased advanced-stage...

ABSTRACT Primary testicular lymphoma (PTL) is a rare occurrence of diffuse large B‐cell (DLBCL) that accounts for 1%–2% all cases. Nodal DLBCL with testis involvement (DLBCL‐T) and PTL are associated poor prognosis, high incidence central nervous system relapse. Fifteen patients (median age 60 years) ( n = 5) or DLBCL‐T 10) received high‐dose methotrexate + R‐CHOP. Overall, complete response (CR) rate was 73% overall 86%. With 3.9‐year median follow‐up, 100% had CR none relapsed. On the...

Extranodal NK/T-cell lymphoma (ENKTCL) is an Epstein-Barr virus (EBV)-related neoplasm preferentially involving the upper aerodigestive tract. Here we show that NK-cell-specific Trp53 disruption in mice leads to development of NK-cell lymphomas after long latency, which involve not only hematopoietic system but also salivary glands. Before tumor onset, knockout causes extensive gene expression changes, resulting immature expansion, exclusively Both human and murine express tissue-resident...

Background: COVID-19 treatment strategies for haematological malignancy patients are adapted from immunocompetent population, without solid evidence. We aimed to describe the efficacy of dexamethasone in a large cohort this population. Methods: Retrospective study multicentre EPICOVIDEHA registry on patients, diagnosed between May 2021 and January 2023. Patients receiving only were compared those plus antiviral strategies. A Cox regression was modelled detect factors associated with...

Extranodal NK/T cell lymphoma (ENKTCL) is a rare subset of peripheral T which Asparaginase containing regimens are the standard care. Despite recent improvements in therapeutic strategy, patient's prognosis remains poor, especially for those who experienced relapsed or refractory (R/R) disease. Recent studies highlighted that immune escape mechanisms involved ENKTCL pathogenesis. Particularly, frequent PD-L1 upregulation led to assess efficacy anti-PD1 (aPD1) therapy small cohorts R/R...
