Severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) utilises the angiotensin-converting enzyme (ACE2) transmembrane peptidase as cellular entry receptor. However, whether SARS-CoV-2 in alveolar compartment is strictly ACE2-dependent and to what extent virus-induced tissue damage and/or direct immune activation determines early pathogenesis still elusive.Spectral microscopy, single-cell/-nucleus RNA sequencing or ACE2 "gain-of-function" experiments were applied infected human lung...

Data are missing on the diversity of Plasmodium spp. infecting apes that live in their natural habitat, with limited possibility human-mosquito-ape exchange. We surveyed wild chimpanzees living an undisturbed tropical rainforest habitat and found 5 species: P. malariae, vivax, ovale, reichenowi, gaboni.

<title>Abstract</title> Lung cancer, the leading cause of cancer-related mortality, presents major challenges for both standard-of-care therapies and innovative treatments like CAR T cells due to tumor heterogeneity resistance. To aid in personalized treatment decisions, preclinical models that incorporate these patient-specific factors are essential. In this study, we developed a platform using matched lung tumoroids healthy organoids. The retained identity original tumors, as confirmed by...

Abstract Severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) primarily targets ciliated cells during the initial infection of upper tract. Since uncertainties persist regarding other involved epithelial cell types, we here utilized viral replication analysis, single-cell RNA sequencing, and spectral microscopy on infected air-liquid interface cultures human primary nasal bronchial to discern type proportions in relation SARS-CoV-2 tropism immune activation. We revealed that, next...

Abstract Mechanisms of epithelial renewal in the alveolar compartment remain incompletely understood. To this end, we aimed to characterize progenitors. Single-cell RNA-sequencing (scRNA-seq) analysis HTII-280 + /EpCAM population from adult human lung revealed subclusters enriched for stem cell signature (ASCS) genes. We found that progenitors organoid culture vitro show phenotypic lineage plasticity as they can yield or bronchial cell-type progeny. The direction differentiation is dependent...

Skin equivalents are increasingly used as human-based test systems for basic and preclinical research. Most of the established skin composed primary keratinocytes fibroblasts, isolated either from excised human or juvenile foreskin following circumcisions. Although potential hair follicle-derived cells generation has been shown, this approach normally requires microdissections scalp which there is limited subject compliance ethical approval. In present study, we report a novel method to...

Streptococcus pneumoniae ( S.p. ) is the most common causative agent of community-acquired pneumonia worldwide. A key pathogenic mechanism that exacerbates severity disease disruption alveolar-capillary barrier. However, specific virulence mechanisms responsible for this in human lung are not yet fully understood. In study, we infected living tissue with and observed a significant degradation central junctional proteins occludin VE-cadherin, indicating barrier disruption. Surprisingly,...

Human-based organ models can provide strong predictive value to investigate the tropism, virulence, and replication kinetics of viral pathogens. Currently, such have received widespread attention in study SARS-CoV-2 causing COVID-19 pandemic. Applicable a large set organoid viruses, we step-by-step work instruction for infection human alveolar-like organoids with this protocol collection.
