Purpose Circulating tumor cells (CTCs) may have utility as surrogate biomarkers and “virtual” biopsies. We report the clinical significance molecular characteristics of CTCs CTC clusters, termed circulating microemboli (CTM), detected in patients with small-cell lung cancer (SCLC) undergoing standard treatment. Patients Methods Serial blood samples from 97 receiving chemotherapy were analyzed using EpCam-based immunomagnetic detection a filtration-based technique. Proliferation status (Ki67)...

Abstract Background With the advent of immunotherapy, substantial progress has been made in improving outcomes for patients with advanced cancer. However, not all benefit equally from treatment, and confounding immune-related issues may have an impact. Several studies suggest that antibiotic use (which alters gut microbiome) result poorer treated immune checkpoint inhibitors (ICI). Materials Methods This is a large, single-site retrospective review n = 291 cancer ICI (n 179 melanoma, 64...

Tumor recurrence after surgical resection of NSCLC obstructs long-term disease-free survival in approximately 50% cases. Our data suggest that combining circulating tumor cell enumeration (as single cells or clusters) tumor-draining pulmonary vein and peripheral blood (assessed by CellSearch) at the time surgery better identifies those patients higher risk for lung cancer than does number alone.

A mass spectrometry-based plasma biomarker discovery workflow was developed to facilitate discovery. Plasma from either healthy volunteers or patients with pancreatic cancer 8-plex iTRAQ labeled, fractionated by 2-dimensional reversed phase chromatography and subjected MALDI ToF/ToF spectrometry. Data were processed using a q-value based statistical approach maximize protein quantification identification. Technical (between duplicate samples) biological variance within individuals)...

Abstract Oncological use of anti-angiogenic VEGF inhibitors has been limited by the lack informative biomarkers. Previously we reported circulating Tie2 as a vascular response biomarker for bevacizumab-treated ovarian cancer patients. Using advanced MRI and biomarkers have extended these findings in metastatic colorectal ( n = 70). Bevacizumab (10 mg/kg) was administered to elicit response, followed FOLFOX6-bevacizumab until disease progression. induced correlation between tumor imaging...

Abstract Serial biopsy of pancreatic ductal adenocarcinoma (PDAC), to chart tumour evolution presents a significant challenge. We examined the utility circulating free DNA (cfDNA) as minimally invasive approach across cohort 55 treatment-naïve patients with PDAC; 31 metastatic and 24 locally advanced disease. Somatic mutations in cfDNA were detected using next generation sequencing 15/24 (62.5%) 27/31 (87%) disease, respectively. Copy number changes 10 whom 7 exhibited gain chromosome 12p...

Vasculogenic mimicry (VM), the process of tumor cell transdifferentiation to endow endothelial-like characteristics supporting de novo vessel formation, is associated with poor prognosis in several types, including SCLC. In genetically engineered mouse models (GEMMs) SCLC, NOTCH, and MYC co-operate drive a neuroendocrine (NE) non-NE phenotypic switch, co-operation between NE cells required for metastasis. Here, we define phenotype VM-competent molecular mechanisms underpinning SCLC VM using...
