Immortal spheroids were generated from fetal mouse intestine using the culture system initially developed to organoids adult intestinal epithelium. Spheroid proportion progressively decreases postnatal period, with a corresponding increase in production of organoids. Like organoids, show Wnt-dependent indefinite self-renewing properties but display poorly differentiated phenotype reminiscent incompletely caudalized progenitors. The spheroid transcriptome is strikingly different that stem...

Apolipoprotein L-I is the trypanolytic factor of human serum. Here we show that this protein contains a membrane pore-forming domain functionally similar to bacterial colicins, flanked by membrane-addressing domain. In lipid bilayer membranes, apolipoprotein formed anion channels. Trypanosoma brucei, was targeted lysosomal and triggered depolarization membrane, continuous influx chloride, subsequent osmotic swelling lysosome until trypanosome lysed.

Multiple phosphatidylinositol (PtdIns) 3-kinases (PI3Ks) can produce PtdIns3P to control endocytic trafficking, but whether enzyme specialization occurs in defined subcellular locations is unclear. Here, we report that PI3K-C2α enriched the pericentriolar recycling compartment (PRE) at base of primary cilium, where it regulates production a specific pool PtdIns3P. Loss PI3K-C2α-derived leads mislocalization PRE markers such as TfR and Rab11, reduces Rab11 activation, blocks accumulation Rab8...

Tsetse flies are the sole vectors of Trypanosoma brucei parasites that cause sleeping sickness. Our knowledge on early interface between infective metacyclic forms and mammalian host skin is currently highly limited. Glossina morsitans infected with fluorescently tagged T. were used in this study to initiate natural infections mice. Metacyclic trypanosomes found be infectious through intradermal route sharp contrast blood stream form trypanosomes. Parasite emigration from dermal inoculation...

Abstract Humans resist infection by the African parasite Trypanosoma brucei owing to trypanolytic activity of serum apolipoprotein L1 (APOL1). Following uptake endocytosis in parasite, APOL1 forms pores endolysosomal membranes and triggers lysosome swelling. Here we show that induces both lysosomal mitochondrial membrane permeabilization (LMP MMP). Trypanolysis coincides with MMP consecutive release Tb EndoG endonuclease nucleus. is associated kinesin KIFC1, which motor vesicular trafficking...

The gain of such organotypic vessel specializations is often deemed to be molecularly unrelated the process organ vascularization. Here, opposing this model, we reveal a molecular mechanism for brain-specific angiogenesis that operates under control Wnt7a/b ligands—well-known blood–brain barrier maturation signals 3–5 relies on Wnt7a/b-dependent expression Mmp25, which find enriched in brain endothelial cells. Progress in diagnosis, treatment, and epidemiology of human African trypanosomiasis (sleeping sickness) depends on the existence specific sensitive diagnostic tools. Inherent shortcomings serologic parasitologic methods can be overcome by molecular techniques. Therefore, we have developed a new polymerase chain reaction (PCR) test using primers derived from recently identified sequence Trypanosoma brucei gambiense-specific glycoprotein (TgsGP). The African trypanosome Trypanosoma brucei, which persists within the bloodstream of mammalian host, has evolved potent mechanisms for immune evasion. Specifically, antigenic variation variant-specific surface glycoprotein (VSG) and a highly active endocytosis recycling coat efficiently delay killing mediated by anti-VSG antibodies. Consequently, conventional VSG-specific intact immunoglobulins are non-trypanocidal in absence complement. In sharp contrast, monovalent antigen-binding...
