Abstract We have previously shown that topotecan, a topoisomerase I poison, inhibits hypoxia-inducible factor (HIF)-1α protein accumulation by DNA damage-independent mechanism. Here, we report daily administration of topotecan HIF-1α expression in U251-HRE glioblastoma xenografts. Concomitant with inhibition, caused significant tumor growth inhibition associated marked decrease angiogenesis and HIF-1 target genes tissue. These results provide compelling rationale for testing clinical trials...

Formalin-fixed, paraffin-embedded (FFPE) tissue is the most common specimen available for molecular assays on after diagnostic histopathological examination. RNA from FFPE suffers strand breakage and cross-linking. Despite excellent extraction methods, quality material remains variable. To address factors within tissues, we studied quality, isolating individual elements of fixation processing including length in formalin type buffer incorporated fixative. We examined impact cycle as well....

Abstract Pancreatic ductal adenocarcinoma (PDAC) is characterized by an abundant desmoplastic stroma. Interactions between cancer and stromal cells play a critical role in tumour invasion, metastasis chemoresistance. Therefore, we hypothesized that gene expression profile of the components pancreatic carcinoma different from chronic pancreatitis reflects interaction with tumour. We investigated eleven tissues PDAC, nine cell lines origin using Affymetrix U133 GeneChip set. The tissue samples...

Regulation of chromatin is an important aspect controlling promoter activity and gene expression. Posttranslational modifications core histones allow proteins associated with transcription to access chromatin. Closely promoters actively transcribed genes, trimethylation histone H3 at lysine 4 (H3K4me3) a mark set by several protein complexes. Some these complexes contain the trithorax ASH2 combined MLL oncoproteins. We identified human in complex oncoprotein MYC. This finding, together...
